目的 解析紫皮石斛(Dendrobium devonianum Paxton)品种"紫玉"及"翠玉"的叶绿体基因组结构及遗传变异特征,开发可用于鉴定的分子标记,为种质资源保护和产业发展提供支撑.方法 采用Illumina NovaSeq平台对2个品种6个样本进行测序,通过GetOrganelle和CPGAVAS2完成叶绿体基因组的组装与注释;利用mVISTA筛选叶绿体基因组高变区间,设计特异引物,并采用PCR扩增、电泳及测序验证其鉴别效果.结果 "紫玉"和"翠玉"的叶绿体基因组长度分别为157115和156632 bp,均具有典型的四分体结构,注释获得119个基因,GC含量均为37.5%.分别检测到46和41个SSRs位点,以A/T型重复为主.序列变异主要集中于非编码区和LSC/SSC区域,共识别出6个高变区间.基于trnI-CAU-trnL-CAA区间设计的引物D5,在100-137 bp区段通过38 bp InDel变异可实现品种鉴别,扩增成功率达100%.结论 明确了"紫玉"和"翠玉"叶绿体基因组的结构特征与遗传差异,开发的分子标记具备良好的稳定性与鉴别效果,为实现紫皮石斛品种级鉴定、种质资源保护及品种权管理提供了有效的分子工具.

Objective To elucidate the chloroplast genome structure and genetic variation of two Dendrobium devonianum Paxton cultivars called"Ziyu"and"Cuiyu",and develop molecular markers for reliable identification,thereby supporting germplasm conservation and industrial development.Methods Six samples from the two cultivars were sequenced using the Illumina NovaSeq platform,chloroplast genomes were assembled and annotated with GetOrganelle and CPGAVAS2;mVISTA was used to screen the highly variable regions of the chloroplast genome,specific primers were designed,and PCR amplification,electrophoresis and sequencing were employed to verify the identification effect.Results The complete chloroplast genomes of"Ziyu"and"Cuiyu"were 157115 and 156632 bp in length,respectively,both exhibiting a typical quadripartite structure.And each genome contained 119 annotated genes,with a GC content of 37.5%.A total of 46 and 41 SSRs loci were detected,predominantly composed of A/T repeats.Sequence variations were mainly concentrated in non-coding regions and the LSC/SSC regions,resulting in the identification of six highly variable regions.A primer pair(D5)targeting the trnI-CAU-trnL-CAA region enabled cultivar discrimination through a 38 bp InDel within the 100-137 bp segment,achieving a 100%amplification success rate.Conclusion This study clarifies the structural features and genetic differences of the chloroplast genomes of"Ziyu"and"Cuiyu".The developed molecular marker exhibits high stability and identification efficacy.These findings provide a robust molecular tool for D.devonianum cultivar authentication,germplasm conservation,and protection of cultivar rights.
