目的 针对山慈菇种苗形态学鉴定局限性,探索高分辨率分子标记并开发特异性引物,以构建精准的山慈菇种苗鉴定体系.方法 对山慈菇3种基原植物叶绿体基因组进行测序、组装与注释.通过全基因组比对分析序列变异位点,利用最大似然法构建系统发育树以评估鉴别能力,并基于高变异区域设计物种特异引物.结果 3种基原植物叶绿体基因组均呈典型环状四分体结构,长度范围为158456-158612 bp,共注释到131-136个基因.SSR位点丰富,以A/T单核苷酸重复为主;密码子使用显示明显的A/U末端偏好.系统发育分析表明,杜鹃兰属与独蒜兰属构成高支持率的单系群.基因组变异分析显示,rps16、atpF、psbH等基因及accD-psal、atpB-rbcL、psbM-trnD-GUC等基因间隔区为高变异区域.结论 叶绿体基因组可高效、精准地鉴别山慈菇基原植物,基于高变区设计的6对特异性引物为其分子鉴定提供了候选工具.本研究为山慈菇种苗鉴定体系的建立奠定了分子基础,对规范药材市场、保障中药质量与用药安全具有重要意义.

Objective To establish a robust molecular foundation for accurate seedling identification,this study addresses the limitations of traditional morphological methods for identifying seedlings of Cremastrae Pseudobulbus Pleiones Pseudobulbus(CPPP)and explores the application of high-resolution molecular markers.Methods The chloroplast genomes of three species of CPPP were sequenced,assembled,and annotated.Subsequently,comparative whole-genome analyses were conducted to detect sequence variation loci.A maximum likelihood phylogenetic tree was constructed to assess the discriminatory power,and specific primers were designed based on hypervariable regions.Results The chloroplast genomes of the three species exhibited a typical circular quadripartite structure,with lengths ranging from 158456 to 158612 bp,and a total of 131 to 136 genes were annotated.Simple sequence repeat(SSR)loci were abundant and primarily consisting of A/T mononucleotide repeats.Codon usage exhibited a significant preference for A/U terminal bases.Phylogenetic analysis revealed that the genus Cremastra and Pleione formed a highly supported monophyletic group.Genome variation analysis identified the coding regions rps16,atpF,psbH,as well as intergenic regions accD-psaI,atpB-rbcL,and psbM-trnD-GUC,as highly variable.Conclusion Chloroplast genomes provide an efficient and accurate method for identifying the original plants of CPPP.The six pairs of specific primers developed in this study offer candidate tools for molecular identification.This research has laid a molecular foundation for the establishment of the ginseng seedling identification system,and is of great significance for standardizing the medicinal material market and ensuring the quality and safety of traditional Chinese medicine.
