不同产地晾制后雪茄烟叶的代谢组差异分析OA
Analysis of Metabolomic Differences in Air-Cured Cigar Tobacco Leaves from Different Production Areas
为阐明不同产区对雪茄烟叶代谢特征及农艺性状的影响,选择四川什邡、山东临朐2个种植产区,选取同一遗传背景下的5个雪茄烟品系QX102、QX105、QX107、QX200、QX205,采用非靶向衍生化GC-MS代谢组学技术结合正交偏最小二乘法辨别分析(OPLS-DA)模型,系统分析两地烟叶代谢组差异及关键代谢通路.结果表明,烟叶中共鉴定出192种代谢物,其中,44种呈现显著产地特异性,四川产区尿嘧啶、酪胺及磷酸三甲基硅烷醇的积累量分别高于山东17.59倍、14.22倍和5.00倍;差异代谢物主要富集于氨基酸代谢和碳氮平衡通路.农艺性状分析结果表明,山东产区烟叶株高、茎围和叶片数分别为221.01、8.59 cm和22.01片,均显著优于四川,且群体表型稳定性更高,其代谢调控网络与氮代谢关键基因的上调表达密切相关.QX105品系在两地均表现代谢组特异性,四川种植的QX105中尿嘧啶异常积累,而山东种植的QX105中单萜合酶活性较高,驱动植醇等萜类物质合成,凸显基因型与环境互作对代谢网络的调控潜力.综上,阐明了不同生态环境下雪茄烟叶代谢物积累与农艺性状的协同响应机制,证实了山东产区更利于高产优质烟叶形成,验证了代谢组学辅助品种适应性评价的可行性,建议以山东临朐为中心示范区扩大雪茄烟种植规模,优先推广QX105等高适应性品系.
To clarify the impact of different production areas on the metabolic characteristics and agronomic traits of cigar tobacco,in this study,the Shifang area in Sichuan and the Linqu area in Shandong were taken as the research objects,five cigar tobacco lines(QX102,QX105,QX107,QX200,and QX205)with the same genetic background were selected.The non-targeted derivatization GC-MS metabolomics technology combined with the orthogonal partial least squares discriminant analysis(OPLS-DA)model was used to systematically analyze the differences in the metabolome of tobacco leaves in the two places and the key metabolic pathways.The results showed that a total of 192 metabolites were identified from tobacco leaves,among which 44 showed significant regional specificity of the production areas.The accumulation amounts of uracil,tyramine,and trimethylsilyl phosphate in the Sichuan production area were 17.59 times,14.22 times,and 5 times higher than those in Shandong,respectively.The differential metabolites were mainly enriched in the amino acid metabolism and carbon-nitrogen balance pathways.The analysis of agronomic traits showed that the plant height 221.01 cm,stem circumference 8.59 cm,and the number of leaves of tobacco in the Shandong production area were significantly better than those in Sichuan,and the population phenotypic stability was higher.Its metabolic regulation network was closely related to the upregulated expression of key genes in nitrogen metabolism.The QX105 line showed metabolomic specificity in both places:uracil accumulated abnormally in QX105 planted in Sichuan,while the monoterpene synthase activity was relatively high in QX105 planted in Shandong,driving the synthesis of terpenoids such as phytol,highlighting the regulatory potential of the interaction between genotype and environment on the metabolic network.In conclusion,the synergistic response mechanism of metabolite accumulation and agronomic traits of cigar tobacco under different ecological environments was clarified,it was confirmed that the Shandong production area was more conducive to the formation of high-yield and high-quality tobacco leaves,the feasibility of metabolomics-assisted variety adaptability evaluation was verified,and it was suggested that the planting scale of cigar tobacco should be expanded with Linqu in Shandong as the central demonstration area and promoting of high adaptability product lines such as QX105 should be prioritized.
