本研究旨在从牛初乳中分离纯化免疫球蛋白M(immunoglobulin M,IgM),并作为抗原免疫BALB/c小鼠,制备鼠抗牛 IgM 多克隆抗体.将牛初乳离心脱脂和盐析后,采用 Sephacryl S-300 凝胶层析与 DEAE-Sepharose 4FF 离子交换层析结合,对牛初乳中 IgM 分离纯化,利用 SDS-PAGE 对得到的高纯度 IgM 进行鉴定,并将纯化后的 IgM 作为抗原,通过免疫注射的方式,制备鼠抗牛 IgM 多克隆抗体,利用 ELISA、Western blot等方法对其抗体进行效价测定和鉴定.结果表明,分离纯化的 IgM的重链分子量为 87.977±0.997 kDa,轻链分子量为25.952±0.160 kDa,纯度达 94%以上,ELISA 结果显示多克隆抗体的效价为 1∶512 000,Western blot 显示单一特异性条带,说明该多克隆抗体与牛 IgM 蛋白发生特异性反应,证明该抗体具有特异性.本研究成功从牛初乳中分离纯化了高纯度的牛初乳 IgM,获得了高效价的鼠抗牛 IgM 多克隆抗体.

This study aims to isolate and purify immunoglobulin M(IgM)from bovine colostrum and uses it as an antigen to immunize BALB/c mice for the preparation of mouse anti-bovine IgM polyclonal antibodies.After centrifugal degreasing and salting-out of bovine colostrum,IgM is isolated and purified by a combination of Sephacryl S-300 gel chromatography and DEAE-Sepharose 4FF ion-exchange chromatography.The obtained high-purity IgM is identified by SDS-PAGE.Subsequently,the purified IgM is used as an antigen to prepare mouse anti-bovine IgM polyclonal antibodies via immunization,and the titer and specificity of the antibodies are determined by ELISA,Western blot,and other methods.The results show that the molecular weights of the heavy and light chains of the isolated and purified IgM are 87.977±0.997 kDa and 25.952±0.160 kDa,respectively,with a purity of over 94%.The titer of the polyclonal antibody is 1∶512 000 as determined by ELISA,and a single specific band is observed in Western blot,indicating that the polyclonal antibody specifically reacts with bovine IgM and has good specificity.This study successfully isolates and purifies high-purity IgM from bovine colostrum and obtains mouse anti-bovine IgM polyclonal antibodies with high titer.
