目的 探究姜黄素(Curcumin)对脂多糖(lipopolysaccharide,LPS)诱导的骨关节炎症损伤的保护作用及其分子机制.方法 体外培养原代小鼠关节软骨细胞,使用不同浓度姜黄素(10、20、40 µmol/L)预处理后,加入LPS构建炎症损伤模型.采用酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测细胞上清液中炎症因子IL-1β、IL-18和TNF-α含量;Western blot分析TXNIP、NLRP3和Caspase-1蛋白表达水平;共聚焦显微镜观察ASC斑点数量以及细胞凋亡率;采用碘乙酸钠(MIA)诱导构建小鼠骨关节炎模型,通过实时荧光定量PCR评估姜黄素对关节软骨的保护作用.结果 姜黄素呈剂量依赖性抑制LPS诱导的炎症因子分泌.机制研究表明,姜黄素可通过与TXNIP蛋白结合,抑制NLRP3炎症小体的活化,进而减少ASC斑点形成和软骨细胞凋亡.体内实验表明,腹腔注射姜黄素20 mg/kg可缓解骨关节炎小鼠膝骨关节炎症反应.结论 姜黄素可通过抑制TXNIP-NLRP3信号通路激活,减轻骨关节炎症反应和软骨细胞凋亡,可能为骨关节炎治疗提供新的潜在药物干预靶点.

Objective This study aimed to investigate the protective effects of curcumin on lipopolysaccharide(LPS)-induced osteoarthritic inflammatory damage and its underlying molecular mechanisms.Methods Primary murine articular chondrocytes were cultured in vitro and pretreated with varying concentrations of curcumin(10,20,and 40 µmol/L)before LPS stimulaltion to establish an inflammation-induced injury model.The levels of inflammatory cytokines IL-1β,IL-18,and TNF-α in the cell supernatant were measured using enzyme-linked immunosorbent assay(ELISA).Western blot analysis was performed to assess the protein expression levels of TXNIP,NLRP3,and Caspase-1.The number of ASC specks and the apoptosis rate of chondrocytes were observed using confocal microscopy.Additionally,a murine osteoarthritis model was established via sodium iodoacetate(MIA)induction,and real-time quantitative PCR(qPCR)was utilized to evaluate the protective effects of curcumin on articular cartilage.Results Curcumin inhibited LPS-induced inflammatory cytokine secretion in a dose-dependent manner.Mechanistically,curcumin was found to bind to TXNIP,thereby suppressing the activation of the NLRP3 inflammasome,leading to a reduction in ASC speck formation and chondrocyte apoptosis.In vivo experiments demonstrated that intraperitoneal administration of curcumin(20 mg/kg)alleviated knee joint inflammation in osteoarthritic mice.Conclusion Curcumin mitigates osteoarthritic inflammation and chondrocyte apoptosis by inhibiting the activation of the TXNIP-NLRP3 signaling pathway,suggesting its potential as a therapeutic target for osteoarthritis treatment.
