目的 优选丹香葡萄糖滴注液中丹参提取液的醇沉工艺.方法 采用Plackett-Burman设计,以水提液密度、第二次醇沉含醇量、pH值作为变量,以丹参素钠、原儿茶醛转移率的综合评分为评价指标,用Box-Behnken响应面法优选醇沉工艺参数.结果 相对密度为 1.15 的丹参水提浓缩液,乙醇醇沉两次,第一次加 95%乙醇至 70%,第二次用 10%氢氧化钠溶液调节pH值至 7.6,再加 95%乙醇至77%,每次在4℃冷藏12 h.测得丹参素钠、原儿茶醛平均转移率分别为58.66%、78.27%,平均综合评分为68.46.结论 优选的醇沉工艺方法稳定可靠,此方法可用于丹香葡萄糖滴注液的醇沉工艺.

Objective To optimize the alcohol precipitation process of Salvia miltiorrhiza extract in Danx-iang Glucose infusion.Methods Plackett-Burman design was employed,with the density of water extract,al-cohol concentration during the second alcohol precipitation,and pH value as variables.The comprehensive score of the transfer rates of salvianic acid A sodium and protocatechuic aldehyde was used as the evaluation index.Box-Behnken response surface methodology was applied to optimize the parameters of alcohol precipitation.Re-sults The concentrated aqueous extract of Salvia miltiorrhiza with a relative density of 1.15 was subjected to al-cohol precipitation twice.Firstly,95%ethanol was added to reach 70%alcohol concentration.Secondly,the pH value was adjusted to 7.6 with 10%sodium hydroxide solution,followed by adding 95%ethanol to 77%alcohol concentration.Each precipitation step was stored at 4℃for 12 h under refrigeration.The average transfer rates of salvianic acid A sodium and protocatechuic aldehyde were 58.66%and 78.27%respectively,with an average comprehensive score of 68.46.Conclusions The optimized alcohol precipitation process is stable and reliable,and it can be applied to the alcohol precipitation procedure of Danxiang Glucose Infusion.
