线粒体自噬在糖尿病周围神经病变中的作用OA
Role of mitophagy in diabetic peripheral neuropathy
糖尿病周围神经病变(diabetic peripheral neuropathy,DPN)是糖尿病最常见的慢性并发症之一,严重影响患者生活质量.其发病机制复杂,涉及代谢紊乱、氧化应激和线粒体功能障碍等多种病理过程.近年来,线粒体自噬作为一种选择性清除受损线粒体的关键细胞质量控制机制,在DPN中受到日益关注.研究表明,线粒体自噬可通过维持线粒体稳态,减轻氧化应激和炎症反应,对DPN的进展起到调节作用.目前线粒体自噬在DPN中的具体分子机制及潜在治疗价值仍不清楚,有待深入研究.系统总结线粒体自噬的分子调控网络,深入探讨其在DPN发生、发展中的作用,并分析靶向线粒体自噬的治疗策略及临床应用前景,有望为理解DPN的发病机制和开发创新治疗方案提供理论依据和研究思路.
Diabetic peripheral neuropathy(DPN)is one of the most common chronic complications of diabetes and seriously affects patients'quality of life.Its pathogenesis is complex and involves multiple pathological processes,including metabolic disorders,oxidative stress,and mitochondrial dysfunction.In recent years,mitophagy,as a key cellular quality-control mechanism for the selective removal of damaged mitochondria,has received increasing attention in DPN.Studies have shown that mitophagy can regulate the progression of DPN by maintaining mitochondrial homeostasis and alleviating oxidative stress and inflammatory responses.At present,the specific molecular mechanisms and potential therapeutic value of mitophagy in DPN remain unclear and require further in-depth investigation.A systematic summary of the molecular regulatory network of mitophagy,an in-depth discussion of its role in the occurrence and development of DPN,and an analysis of therapeutic strategies targeting mitophagy and their clinical application prospects are expected to provide a theoretical basis and research ideas for understanding the pathogenesis of DPN and developing innovative treatment strategies.
