目的 探讨桔梗多糖对溃疡性结肠炎(ulcerative colitis,UC)大鼠的氧化应激反应及Toll样受体(Toll-like receptor,TLR)/核转录因子 κB(nuclear transcription factor kappa B,NF-κB)、Janus 激酶 2(Janus kinase 2,JAK2)/信号转导与转录激活子 3(signal transduction and activator of transcription 3,STAT3)信号通路相关因子表达的影响.方法 葡聚糖硫酸钠(dextran sulfate sodium,DSS)建立UC大鼠模型并给予不同剂量桔梗多糖处理 2 周,观察大鼠疾病活动指数(disease activity index,DAI)、结肠病理改变、细胞凋亡状况、炎症和氧化应激因子水平、TLR4/NF-κB和JAK2/STAT3 相关mRNA和蛋白质表达.结果 桔梗多糖处理可以降低结肠损伤严重程度,结肠组织细胞凋亡率,IL-6、TNF-α、IL-17、MDA水平,升高IL-10、SOD和GSH-px水平,降低结肠组织TLR4、NF-κB、JAK2、STAT3,以及p-JAK2、p-STAT3 的表达,且呈现剂量依赖效应(P<0.05).结论 桔梗多糖可以改善UC大鼠结肠损伤,缓解结肠组织氧化应激,还对TLR/NF-κB、JAK2/STAT3 通路激活有抑制作用.

Objective To explore the effect of Platycodon grandiflorum polysaccharides on oxi-dative stress,and expression of Toll-like receptor(TLR)/nuclear transcription factor kappa B(NF-κB)and Janus kinase 2(JAK2)/signal transduction and activator of transcription 3(STAT3)signaling pathways-related factors in rats with ulcerative colitis(UC).Methods The UC rat model was constructed by dextran sulfate sodium(DSS),then UC rats were treated with different doses of Platycodon grandiflorum polysaccharides for 2 weeks.The disease activity index,pathological changes of colon,apoptosis,levels of inflammatory and oxidative stress factors,and expression levels of TLR4/NF-κB and JAK2/STAT3 related mRNA and proteins were ob-served.Results After treatment with Platycodon grandiflorum polysaccharides,the disease activity index,apoptosis rate of colon tissues,levels of IL-6,TNF-α,IL-17 and MDA were decreased,levels of IL-10,SOD and GSH-px were increased,expression levels of TLR4,NF-κB,JAK2,STAT3,and p-JAK2 and p-STAT3 in colon tissues were decreased,with a dose dependent manner(P<0.05).Conclusion Platycodon grandiflorum polysaccharides can alleviate colon injury and oxidative stress in colon tissues in UC rats,and inhibit the activation of TLR/NF-κB and JAK2/STAT3 pathways.
