农杆菌介导剑叶文心兰遗传转化体系中抗生素的筛选OA

Screening of antibiotics in the Agrobacterium-mediated genetic transformation system of leaf disc method for Tolumnia

为完善剑叶文心兰高效稳定的遗传转化体系,以离体叶片为材料,通过浓度梯度试验与数理统计分析,探究 3 种选择抗生素对愈伤组织的影响及 2 种抑菌抗生素的效果.结果显示,经Duncan多重比较与方差分析,Basta 0.18 mg/L、Hyg 9 mg/L、Kana 100 mg/L可完全抑制叶片不定芽分化(P<0.01),作为抗性芽选择压;Basta 0.12 mg/L、Hyg 5 mg/L、Kana 40 mg/L为继代苗半致死浓度(P<0.05),作为抗性苗筛选浓度.剑叶文心兰对抗生素敏感性为Basta>Hyg>Kana(P<0.01),更适合选用bar基因作为选择标记.100 mg/L Cef可有效抑制农杆菌且叶片存活率高.研究结果为剑叶文心兰遗传转化体系优化及转基因植株筛选提供了关键参数.

To optimize the efficient and stable genetic transformation system of Oncidium ensifolium,in vitro leaves were used as explants in this study.The effects of 3 types of selective antibiotics on callus and the efficacy of 2 types of bacteriostatic antibiotics were investigated via concentration gradient assays and mathematical/statistical analyses.Results showed that,as revealed by Duncan's multiple range test and analysis of variance(ANOVA),0.18 mg/L Basta,9 mg/L hygromycin(Hyg),and 100 mg/L kanamycin(Kana)completely inhibited adventitious bud differentiation in leaves(P<0.01),and thus were identified as the selection pressure for resistant buds.The half-lethal concentrations(LC50)for subcultured seedlings were 0.12 mg/L Basta,5 mg/L Hyg,and 40 mg/L Kana(P<0.05),which were designated as the screening concentrations for resistant seedlings.O.ensifolium exhibited antibiotic sensitivity in the order of Basta>Hyg>Kana(all P<0.01),indicating that the bar gene is more suitable as a selectable marker.Additionally,100 mg/L cefotaxime(Cef)effectively inhibited Agrobacterium proliferation while maintaining a high leaf survival rate.This study provides critical parameters for optimizing the genetic transformation system and screening transgenic plants in Tolumnia.
