猪圆环病毒2型去核定位信号Cap蛋白的原核表达及多克隆抗体制备OA
Prokaryotic expression of Capsid Protein without nuclear localization signal of Porcine circovirus 2 and preparation for its polyclonal antibody
为制备猪圆环病毒 2 型(Porcine Circovirus type 2,PCV2)衣壳蛋白(Capsid Protein,Cap)的多克隆抗体,以pMD18-T-ORF2 质粒为模板,扩增去核定位信号序列(NLS)的Cap蛋白基因片段,并将其克隆至原核表达载体pET-28a(+)中,构建重组表达质粒pET-28a-N-Cap.将该重组质粒转化至大肠杆菌Rosetta(DE3)感受态细胞中,经IPTG诱导表达后,SDS-PAGE分析显示重组蛋白主要以不可溶性形式存在.利用镍柱对重组蛋白进行纯化,并免疫兔制备兔源多克隆抗体.结果显示,所制备的多克隆抗体效价可达1∶204 800.经IFA和Western blot分析,该多克隆抗体能特异性识别PCV2.结果表明,经原核系统表达的去核定位信号的PCV2 Cap蛋白具有良好的免疫原性,且所获得的多克隆抗体可用于Cap蛋白的相关检测.
To generate polyclonal antibodies targeting the Cap protein of Porcine Circovirus type 2(PCV2),a Cap gene fragment without the nuclear localization signal(NLS)was amplified using the pMD18-T-ORF2 plasmid as a template.This fragment was subsequently cloned into the prokaryotic expression vector pET-28a(+)to construct the recombinant plasmid pET-28a-N-Cap.The recombinant plasmid was then transformed into Escherichia coli Rosetta(DE3)competent cells.After induction with IPTG,SDS-PAGE analysis revealed that the expressed recombinant protein was predominantly present in an insoluble form.The target protein was purified via nickel-affinity chromatography and used as an antigen to immunize rabbits for the production of rabbit-derived polyclonal antibodies.The resulting antibody titer reached up to 1∶204,800,as determined by serological testing.Furthermore,indirect immunofluorescence assay(IFA)and Western blot analysis confirmed that the obtained polyclonal antibodies specifically recognized PCV2.These findings indicate that the NLS-deleted Cap protein expressed in a prokaryotic system retains strong immunogenicity,and the acquired polyclonal antibodies can be utilized in assays related to the Cap protein.
