湖北恩施野生茶树种质资源生化成分多样性分析OA
Diverse Biochemistry of Wild Tea Germplasms in Enshi,Hubei
[目的]探明湖北恩施野生茶树种质资源生化成分特点,为野生茶树进一步开发利用提供依据.[方法]采用高效液相色谱(high performance liquid,HPLC)等方法,对湖北省恩施州 5个县市共 70份野生茶树样本的儿茶素组分、咖啡碱、氨基酸含量进行检测,并运用主成分分析和聚类分析等方法,对其生化成分多样性进行分析和综合评价.[结果]野生茶树生化成分多样性指数(index of genetic diversity,H')为 1.50~3.09,平均值为 2.41.变异系数(coefficients of variation,CV)为 23.06%~96.47%,平均值为 41.95%.共有 3个主成分因子特征值大于 1,累计贡献率达 80.59%,对 3个主成分贡献最大的分别是表儿茶素没食子酸酯(epicatechingallate,ECG)、表没食子儿茶素(epigallocatechin,EGC)、儿茶素(catechin,C).聚类分析结果表明,70份野生茶树资源可划分为 4个类群.其中,第Ⅰ类群非酯型儿茶素含量较高;第Ⅱ类群儿茶素含量在 4个类群中最低;第Ⅲ类群氨基酸含量在 4个类群中最低;第Ⅳ类群酯型儿茶素含量为 4个类群中最高.样本回归方程计算评分显示,钟台村大茶树 4号、大集场村大茶树1号、钟台村大茶树2号3份样本得分最高且EGC、EGCG、总儿茶素、咖啡碱含量较高,具有特异性.[结论]恩施野生茶树种质资源生化成分变异范围较大,遗传多样性丰富.EGC、EGCG、咖啡碱 3个生化成分可作为湖北恩施野生茶树的综合评价指标.钟台村大茶树 4号、大集场村大茶树 1号和钟台村大茶树 2号 3份资源具有选育优质茶树品种的潜力.
[Objective]Vastly diverse biochemical compositions of wild tea germplasms in Enshi,Hubei were analyzed to facilitate development and utilization of the natural resources.[Method]HPLC and other analytical methods were applied to determine the contents of catechins,caffeine,and amino acids in 70 wild tea cultivars from 5 counties and cities in Enshi Prefecture,Hubei Province.Principal component and cluster analyses were conducted to categorize these germplasms based on biochemical diversity.[Result]The indexes of genetic diversity on biochemistry of the germplasms ranged 1.50–3.09,averaging 2.41,and coefficients of variation 23.06%–96.47%,averaging 41.95%.Of the principal components,epicatechin gallate(ECG),epigallocatechin gallate(EGCG),and catechin(C),with eigenvalues greater than 1 cumulatively contributed 80.59%of total explanation.A cluster analysis grouped the 70 germplasms into 4 categories:Group I of a relatively high content in non-ester catechin,Group II of the lowest average content in catechin,Group III of the lowest in amino acid content,and Group IV of the highest ester catechins content.The cultivars,Zhongtai Village Dacheshan No.4,Dajichang Village Dacheshan No.1,and Zhongtai Village Dacheshan No.2 with high contents of ECG,EGCG,total catechins,and caffeine received the highest scores and were selected as the outstanding varieties by regression analysis.[Conclusion]The biochemicals in the genetically richly divese 70 wild tea germplasms of Enshi varied significantly.ECG,EGCG,and caffeine were the crucial substances contributing to the tea quality and could be used as indicators for comprehensive evaluation.Accordingly,Zhongtai Village Dacheshan No.4,Dajichang Village Dacheshan No.1,and Zhongtai Village Dacheshan No.2 were selected as potential candidates for breeding new varieties.
