Effects of Dietary Coconut Oil Supplementation on Lipopolysaccharide-Induced Muscle Injury and cGAS-STING Signaling Pathway in Piglets

本试验旨在研究饲粮中添加椰子油对脂多糖(LPS)诱导的仔猪肌肉损伤和环鸟苷酸-腺苷酸合成酶(cGAS)-干扰素基因刺激因子(STING)信号通路的影响.选取 28 日龄的"杜×长×大"断奶仔猪 24 头,随机分为 4 个组,每组 6 个重复,每个重复 1 头猪.采用 2×2 因子设计,主因子分别为饲粮处理(添加3%大豆油或3%椰子油)和LPS处理(注射LPS或生理盐水).在试验第 28 天,仔猪腹腔注射 100 μg/kg BW的LPS或等量生理盐水,4 h后屠宰取样.结果表明:1)在LPS刺激下椰子油显著降低了血清中总蛋白(TP)含量(P<0.05),并缓解了LPS诱导的血清中葡萄糖(GLU)含量的降低(P>0.05);2)椰子油显著加剧了LPS诱导的肌肉中白细胞介素-6(IL-6)mRNA相对表达量的升高(P<0.05),并显著抑制了LPS诱导的肌肉中白细胞介素-10(IL-10)mRNA相对表达量的升高(P<0.05);3)椰子油显著降低了肌肉中丙二醛(MDA)含量(P<0.05),且显著缓解了LPS诱导的肌肉中总抗氧化能力(T-AOC)的升高(P<0.05);4)椰子油显著提高了肌肉中叉头框蛋白O1(FOXO1)mRNA相对表达量(P<0.05),并在LPS刺激下显著提高了肌肉中FOXO1 和叉头框蛋白O4(FOXO4)mRNA相对表达量(P<0.05);5)椰子油显著提高了肌肉中 2′,5′-寡腺苷酸合成酶 2(OAS2)mRNA相对表达量(P<0.05),且显著加剧了LPS诱导的肌肉中STING mRNA相对表达量的升高(P<0.05).综合以上结果得出,饲粮中采用椰子油代替等量的大豆油会促进cGAS-STING信号通路的激活,加剧LPS诱导的仔猪肌肉炎症反应和蛋白质降解.

This experiment aimed to investigate the effects of dietary coconut oil supplementation on lipopo-lysaccharide(LPS)-induced muscle injury and cyclic GMP-AMP synthase(cGAS)-stimulator of interferon gene(STING)signaling pathway in piglets.A total of twenty-four 28-day-old"Duroc×Landrace×Yorkshire"weaned piglets were randomly allocated to four groups with six replicates per group and one piglet per replicate.A 2×2 factorial design was adopted,with the main factors being dietary treatment(supplemented with 3%soy-bean oil or 3%coconut oil)and LPS treatment(injected with LPS or saline).On day 28 of the trial,piglets were intraperitoneally injected with 100 μg/kg BW LPS or an equal volume of saline,and samples were col-lected after slaughter 4 hours later.The results showed as follows:1)coconut oil significantly reduced the ser-um total protein(TP)content under LPS challenge(P<0.05),and alleviated the LPS-induced decrease in ser-um glucose(GLU)content(P>0.05).2)Coconut oil significantly exacerbated the LPS-induced increase in the mRNA relative expression level of interleukin-6(IL-6)in muscle(P<0.05),and significantly inhibited the LPS-induced increase in the mRNA relative expression level of interleukin-10(IL-10)in muscle(P<0.05).3)Coconut oil significantly reduced the muscle malondialdehyde(MDA)content(P<0.05)and sig-nificantly alleviated the LPS-induced increase in muscle total antioxidant capacity(T-AOC)(P<0.05).4)Coconut oil significantly increased the mRNA relative expression level of forkhead box protein O1(FOXO1)in muscle(P<0.05),and significantly increased the relative mRNA expression of both FOXO1 and forkhead box protein O4(FOXO4)in muscle under LPS challenge(P<0.05).5)Coconut oil significantly increased the mRNA relative expression level of 2′,5′-oligoadenylate synthetase 2(OAS2)in muscle(P<0.05),and significantly exacerbated the LPS-induced increase in the mRNA relative expression level of STING in muscle(P<0.05).Based on the above results,it can be concluded that replacing an equal amount of soybean oil with coconut oil in the diet promotes the activation of the cGAS-STING signaling pathway,and exacerbates LPS-in-duced muscle inflammation and protein degradation in piglets.[Chinese Journal of Animal Nutrition,2026,38(3):2164-2172]
