火烧法胁迫下棋楠沉香品质及功能活性评价OA

Evaluation of Quality and Functional Activity of Aquilaria sinensis Under the Stress of Fire-Burning Method

以45 s(SW)、60s(LW)和75s(QW)3种火烧时间处理棋楠木以诱导结香,处理12个月后采集棋楠沉香样品,测定其乙醇提取物与精油得率,进行显色反应鉴定,并利用气相色谱-质谱联用技术(GC-MS)分析其化学成分.进一步评估样品对DPPH、ABTS+自由基的清除能力以及抗乙酰胆碱酯酶活性.结果表明:(1)SW、LW和QW 3组处理的乙醇提取物质量分数分别为(54.27±0.89)%、(56.14±1.08)%和(61.37±1.81)%,精油质量分数分别为(41.62±1.02)%、(42.20±1.63)%和(46.87±0.77)%;(2)所有样品显色反应均呈樱红色或紫堇色,符合《中华人民共和国药典》对理化显色反应的要求;(3)共鉴定出89种化合物,各处理组以2-(2-苯乙基)色酮类(PECs)、倍半萜类和芳香族类化合物为主,SW、LW和QW中3类化合物的数量分别为33、43和35个,相对质量分数总和分别为94.73%、95.65%和96.22%,而对照组(CK)以脂肪族类和芳香族类化合物为主;(4)在活性方面,SW、LW和QW对DPPH自由基的半数抑制质量浓度(IC50)分别为0.286 2、0.259 8和0.157 2g/L,对ABTS+自由基的IC5.值分别为0.5490、0.3477和0.1078g/L,对乙酰胆碱酯酶的IC5.值分别为1.9168、1.3375和1.1670g/L.综上,火烧胁迫可显著提高棋楠沉香的结香效率与品质,其中QW(75s)处理在提取物得率、特征成分含量及活性方面表现最佳.为进一步明确火烧时间对沉香品质的临界效应,后续研究可增设更长处理时间(如90、105 s)的实验组,以揭示成分积累的阈值及更优胁迫条件.火烧法是一种高效、优质的结香诱导方式,对推动沉香高质量生产具有重要应用价值.

Aquilaria sinensis was induced by treating A.sinensis with three fire-burning durations,namely 45 s(SW),60 s(LW),and 75 s(QW).After 12 months of treatment,A.sinensis samples were collected to determine the yields of ethanol extracts and essential oils,conduct color reaction identification,and analyze their chemical compositions by gas chromatog-raphy-mass spectrometry(GC-MS).Furthermore,the samples'scavenging capacities against DPPH and ABTS+free radi-cals as well as their anti-acetylcholinesterase activity were evaluated.The results showed that:(1)The mass fractions of ethanol extracts in the SW,LW,and QW groups were(54.27±0.89)%,(56.14±1.08)%,and(61.37±1.81)%respec-tively,while the mass fractions of essential oils were(41.62±1.02)%,(42.20±1.63)%,and(46.87±0.77)%respec-tively.(2)All samples exhibited cherry red or violet color in the color reactions,which met the requirements for physico-chemical color reactions specified in the Pharmacopoeia of the People's Republic of China.(3)A total of 89 compounds were identified.The main components in each treatment group were 2-(2-phenylethyl)chromones(PECs),sesquiterpe-noids,and aromatic compounds.The numbers of these three types of compounds in SW,LW,and QW were 33,43,and 35 respectively,with the total relative mass fractions reaching 94.73%,95.65%,and 96.22%respectively.In contrast,the control group(CK)was dominated by aliphatic and aromatic compounds.(4)In terms of bioactivity,the half-maximal inhibitory concentrations(IC50)of SW,LW,and QW against DPPH free radicals were 0.286 2,0.259 8,and 0.157 2 g/L respectively,their IC50 values against ABTS+free radicals were 0.549 0,0.347 7,and 0.107 8 g/L respectively,and their IC50 values against acetylcholinesterase were 1.916 8,1.337 5,and 1.167 0 g/L respectively.In conclusion,fire-burning stress can significantly improve the agarwood-inducing efficiency and quality of A.sinensis,among which the QW(75 s)treatment showed the best performance in extract yield,characteristic component content,and bioactivity.To fur-ther clarify the critical effect of fire-burning duration on agarwood quality,subsequent studies can add experimental groups with longer treatment durations(e.g.,90 s and 105 s)to reveal the threshold of component accumulation and optimal stress conditions.The fire-burning method is an efficient and high-quality agarwood-inducing approach,which has important ap-plication value for promoting the high-quality production of agarwood.
