女贞子多糖对H22荷瘤小鼠的抗肿瘤作用及机制研究OA
Antitumor Effect and Mechanism of Ligustrum lucidum Polysaccharide in H22 Tumor-Bearing Mice
目的:研究女贞子多糖对H22 荷瘤小鼠的抑瘤作用及其作用机制.方法:建立H22 荷瘤小鼠模型,连续给药14 d后,解剖小鼠并称瘤质量,计算抑瘤率;ELISA法检测小鼠血清中肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、和γ-干扰素(IFN-γ)含量;Annexin-V-PE/7AAD双染流式技术检测肿瘤细胞凋亡与自噬蛋白LC-3A/B蛋白的表达水平;Western blot法检测肿瘤组织中促凋亡蛋白(BAX)、抗凋亡蛋白(BCL2)、自噬蛋白标志物LC3Ⅱ/Ⅰ、Beclin1、蛋白激酶B(AKT)、哺乳动物雷帕霉素靶蛋白(mTOR)的表达.结果:女贞子多糖有明显的抑瘤作用,抑瘤率可达到50.83%.与模型对照组相比,女贞子多糖 20 mg/kg小鼠血清中TNF-α、IL-6 与 IFN-γ含量明显降低(P<0.05 或P<0.01),肿瘤细胞凋亡率升高,BAX表达上调,BCL2 表达下调(P<0.05);LC3A/B、LC3Ⅱ/Ⅰ增加、Beclin1 表达上调、AKT与mTOR的蛋白表达下调(P<0.05 或P<0.01).结论:女贞子多糖可抑制H22荷瘤小鼠肿瘤生长,促进瘤组织细胞凋亡,提高肿瘤组织细胞自噬,其机制可能是通过阻滞AKT/mTOR信号通路起到抗肿瘤作用.
Objective:To investigate the antitumor effect and mechanism of Ligustrum lucidum polysaccharide in H22 tumor-bearing mice.Methods:The mouse model of H22 hepatocellular carcinoma was established.After 14 days of continuous administration,mice were dissected and tumor weights were measured to calculate the tumor inhibition rate.Serum levels of tumor necrosis factor(TNF)-α,interleukin(IL)-6,and interferon(IFN)-γ were determined by ELISA.Annexin-V-PE/7AAD dual-staining flow cytometry was employed to assess tumor cell apoptosis and expression of the au-tophagy and apoptosis marker microtubule-associated protein 1 light chain 3 A/B(LC3A/B).Western blot was em-ployed to evaluate the expression levels of the anti-apoptotic protein B-cell lymphoma 2(Bcl2),the pro-apoptotic protein Bcl2-associated X protein(Bax),LC3Ⅱ/Ⅰ,beclin1,protein kinase B(AKT),and mammalian target of rapamycin(mTOR).Results:L.lucidum polysaccharide exhibited significant antitumor activity,achieving a tumor inhibition rate of 50.83%.Compared with the model group,the L.lucidum polysaccharide(20 mg/kg)group showed decline serum levels of TNF-α,IL-6,and IFN-γ(P<0.01 or P<0.05),increased tumor cell apoptosis,up-regulated Bax expression,and down-regulated Bcl2 expression(P<0.05).In addition,this group showed increases in the percentage of LC3A/B and LC3Ⅱ/Ⅰ,and the expression levels of beclin1,reduces in the expression levels of AKT and mTOR(P<0.05,P<0.01).Conclusion:L.lucidum polysaccharide can inhibit the tumor growth and promote the tumor cell apoptosis and auto-phagy in H22 tumor-bearing mice by inhibiting the AKT-mTOR signaling pathway.
