西瓜品种KASP指纹图谱构建和种子纯度鉴定OA
Construction of KASP-Based Molecular Fingerprinting Profiles for Citrullus lanatus Cultivars and Assessment of Its Seed Purity
分子标记技术在植物品种真实性及种子纯度鉴定中具有独特的优势.以西瓜杂交种中农黄冠1号及其父母本为试验材料,筛选竞争性等位基因特异性PCR(KASP)标记用于构建品种DNA指纹图谱和鉴定种子纯度.结果表明,32个核心KASP标记有28个分型成功,根据位点基因型构建了中农黄冠1号DNA指纹图谱,28个标记中有6个能够将中农黄冠1号的父本和母本区分开,选用2个多态性标记WmSNP69和WmSNP89检测杂交种纯度,种子纯度鉴定结果分别为95.17%和95.14%,进一步采用简单重复序列(SSR)标记和田间鉴定结果相符,证明KASP标记用于西瓜杂交种纯度鉴定是可行的.研究构建了中农黄冠1号西瓜品种的单核苷酸多态性(SNP)指纹图谱,并筛选出适合种子纯度鉴定的KASP标记,为品种知识产权保护和种子质量检测提供了技术支撑.
Molecular marker technology has unique advantages in the authenticity verification and seed purity determination of plant varieties.Using the hybrid Citrullus lanatus variety Zhongnonghuang-guan No.1 and its parents as the experimental materials,competitive allele-specific PCR(KASP)markers were screened to construct the DNA fingerprint profiles of the variety and determine the seed purity.The results showed that 28 out of 32 core KASP markers were successfully typed.Based on the genotype of the loci,the Zhongnonghuangguan No.1 DNA fingerprint profiles was constructed.Among the 28 markers,6 could distinguish the father and mother plants of Zhongnonghuangguan No.1.2 polymorphic markers WmSNP69 and WmSNP89 were selected to detect the purity of the hy-brid variety,and the seed purity determination results were 95.17%and 95.14%respectively.Further,the simple repetitive sequence(SSR)markers and field identification results were consist-ent,proving that the KASP markers were feasible for the purity determination of the hybrid variety of Citrullus lanatus.The study constructed the single nucleotide polymorphism(SNP)fingerprint profiles of the Citrullus lanatus variety Zhongnonghuangguan No.1,and selected the KASP markers suitable for seed purity determination,providing technical support for the protection of variety intellectual property rights and seed quality detection.
