百脉根伸展蛋白基因LjEXT1调控根系生长的功能分析OA
Functional Analysis of the Extensin Protein Gene LjEXT1 in Regulating Root Growth of Lotus corniculatus
伸展蛋白在植物细胞形态建成、细胞壁合成和根生长中起重要作用.为探究LjEXT1基因在根系发育中的调控作用,通过生物信息学预测和植物遗传转化方法研究百脉根LjEXT1基因的功能,利用qRT-PCR、GUS组织化学染色及基因组PCR进行基因表达分析和转基因鉴定,采用钙白荧光染色和石蜡切片观察根细胞壁形成和细胞结构,并通过TTC法测定根系活力.结果表明,LjEXT1基因主要在根中表达,在豆科植物中高度保守,与逆境响应和根发育有关;超量表达LjEXT1使根系分化延迟3~6d,抑制根系伸长和不定根形成,根系活力降低至62.50%;组织切片和原生质体观察显示,转基因植株不定根发生部位细胞壁增厚20.42%,根细胞壁形成提前约24 h;基因表达分析发现,LjEXT1影响细胞壁调控相关基因LjEXP、LjRLAF和LjRHS的表达.研究表明,LjEXT1基因负调控根系生长,并可能与促进细胞壁增厚有关.
The extensin protein plays an important role in the formation of plant cell morphology,cell wall syn-thesis and root growth.To investigate the regulatory role of the LjEXT1 gene in root development,the function of the LjEXT1 gene in Lotus corniculatus was studied through bioinformatics prediction and plant genetic trans-formation methods.Gene expression analysis and transgenic identification were conducted using qRT-PCR,GUS histochemical staining and genomic PCR.Calcium white fluorescence staining and paraffin sectioning were used to observe root cell wall formation and cell structure and the root vitality was determined by the TTC method.The results showed that the LjEXT1 gene was mainly expressed in the roots and was highly conserved in Fabaceae plants,related to stress response and root development;overexpression of LjEXT1 delayed root differentiation by 3-6 days,inhibited root elongation and the formation of adventitious roots and reduced root vitality to 62.50%.The tissue sections and protoplast observations revealed that the cell walls of cells at the site of adventi-tious roots in transgenic plants were thickened by 20.42%and root cell wall formation was advanced by approxi-mately 24 hours.The gene expression analysis found that LjEXT1 affected the expression of cell wall regulation-related genes LjEXP,LjRLAF,and LjRHS.The study showed that the LjEXT1 gene negatively regulates root growth and could be related to promoting cell wall thickening.
