miR-363-3p与FZD10在肝细胞癌中的表达及临床病理特征与预后的关系OA
The relationship between the expression of miR-363-3p and FZD10 in hepatocellular carcinoma tissues and the clinical pathological characteristics and prognosis
目的:探讨微小RNA-363-3p(miR-363-3p)与卷曲类受体10(FZD10)在肝细胞癌(HCC)组织中的表达及与临床病理特征和预后的关系.方法:选取2018年1月至2021年1月在南阳市中心医院诊治的86例HCC患者,采集术中切除的癌组织和癌旁组织为实验材料,实时荧光定量聚合酶链反应(qRT-PCR)检测miR-363-3p、FZD10 mRNA水平,免疫组织化学染色法检测FZD10蛋白表达.采用Pearson法分析miR-363-3p、FZD10 mRNA的相关性;通过StarBase网站预测miR-363-3p、FZD10结合位点;Kaplan-Meier进行生存分析,Cox回归分析HCC患者预后不良的影响因素.结果:miR-363-3p在癌旁组织高表达,FZD10 mRNA、FZD10蛋白在癌组织中高表达(P<0.05);miR-363-3p、FZD10 mRNA表达呈负相关(r=-0.455,P<0.001),StarBase网站预测miR-363-3p与FZD10 mRNA之间存在结合位点;癌组织miR-363-3p、FZD10表达与患者微血管侵犯、肝内转移、肝硬化、淋巴结状态相关(P<0.05);miR-363-3p高表达患者累积生存率为78.57%(33/42),高于miR-363-3p低表达的45.45%(20/44);FZD10高表达患者累积生存率为48.83%(22/48),低于FZD10低表达的81.58%(31/38)(P<0.05);miR-363-3p、FZD10、肝内转移、肝硬化、淋巴结状态为HCC患者预后不良的独立影响因素(P<0.05).结论:在HCC组织中,miR-363-3P表达下调而FZD10表达上调,两者表达呈负相关,且均与肿瘤侵袭性病理特征及不良预后密切相关.
Objective:To investigate the expression of microRNA-363-3p(miR-363-3p)and frizzled 10(FZD10)in liver cancer tissue and their relationship with clinical pathological features and prognosis.Methods:This study selected 86 liver cancer patients who were treated in our hospital from January 2018 to January 2021,and the excised cancer tissue and adjacent tissues during surgery were used as experimental materials.Real-time fluorescence quantitative PCR was applied to detect miR-363-3p and FZD10 mRNA levels.Immunohistochemical staining(IHC)was applied to detect FZD10 protein level.Pearson correlation was applied to analyze the correlation between miR-363-3p and FZD10 mRNA.StarBase website was applied to predict the binding sites of miR-363-3p and FZD10.Kaplan-Meier was applied for survival analysis.COX regression was applied to analyze the influencing factors of poor prognosis.Results:miR-363-3p was highly expressed in adjacent cancer tissues,while FZD10 mRNA and FZD10 protein were highly expressed in cancer tissues(P<0.05).The expression of miR-363-3p and FZD10 mRNA was negatively correlated(r=-0.455,P=0.000),and StarBase prediction showed a binding site between miR-363-3p and FZD10 mRNA.The expression of miR-363-3p and FZD10 in cancer tissue was correlated with microvascular invasion,intrahepatic metastasis,cirrhosis,and lymph node status in patients(P<0.05).The cumulative survival rate of patients with high expression of miR-363-3p was 78.57%(33/42),which was higher than 45.45%(20/44)of patients with low expression of miR-363-3p.The cumulative survival rate of patients with high expression of FZD10 was 48.83%(22/48),which was lower than 81.58%(31/38)of patients with low expression of FZD10(P<0.05).miR-363-3p,FZD10,intrahepatic metastasis,cirrhosis,and lymph node status were independent influencing factors for poor prognosis in patients(P<0.05).Conclusion:miR-363-3p is poorly expressed in cancer tissue,while FZD10 mRNA and protein are highly expressed in cancer tissue.Both are associated with clinical pathological features such as microvascular invasion,intrahepatic metastasis,cirrhosis,and lymph node status.Patients with low expression of miR-363-3p and high expression of FZD10 have lower survival rates.
