Analysis on Chemical Component Differences between Cultivated Aquilaria sinensis Qi-Nan and Wild Aquilaria sinensis

为探究人工栽培奇楠沉香和野生沉香特征性成分的差异,通过乙醚超声浸提法提取人工栽培奇楠沉香金丝油、油叶子、金砂叶以及野生沉香样品进行气相色谱-质谱(GC-MS)特征性成分检测分析,并分别采用MTT比色法、滤纸片琼脂扩散法测定沉香乙醚浸提物细胞毒和抑菌活性.结果表明,不同材料间乙醚浸提物得率分化较大;4种沉香样品共检测出25种化学成分,主要为倍半萜和色酮类化合物;与野生沉香相比,人工栽培奇楠沉香色酮类化合物2-(2-苯乙基)色酮和2-[2-4-(甲氧基苯)乙基]色酮相对含量之和较高,但鉴定出的倍半萜种类较少,总相对含量较低;生物活性方面,油叶子乙醚浸提物对慢性髓原白血病细胞(K-562)和宫颈癌细胞(HeLa)表现出较高的抑制活性,金丝油乙醚浸提物对金黄色葡萄球菌的抑制能力较强.表明人工栽培奇楠沉香和野生沉香乙醚浸提物化学成分均以倍半萜和色酮类为主,但在特征性成分含量及活性方面存在差异.

To explore the differences in characteristic components between cultivated Aquilaria sinensis Qi-Nan and wild Aquilaria sinensis,the ethyl ether ultrasonic extraction method was used to extract the Jinsiyou,Youyezi,Jinshaye of cultivated Aquilaria sinensis Qi-Nan,and wild Aquilaria sinensis samples for gas chromatography-mass spectrometry(GC-MS)characteristic component detection and analysis.The cell toxicity and antibacterial activities of the ethyl ether extracts from the Aquilaria sinensis samples were determined using the MTT colorimetric method and filter paper agar diffusion method.The results showed that the yield of ethyl ether extracts varied significantly among different materials.A total of 25 chemical components were detected in the 4 Aquilaria sinensis samples,mainly including sesquiterpenes and ketone compounds.Compared with wild Aquilaria sinensis,the sum of the relative contents of ketone compounds 2-(2-phenylethyl)ketone and 2-[2-4-(methoxy phe-nyl)ethyl]ketone in cultivated Aquilaria sinensis was higher,but the types of sesquiterpenes identified and the total relative content were lower.In terms of biological activity,the ethyl ether extract of Youyezi showed higher inhibitory activity against human chronic myeloid leukemia cells(K-562)and human cervical cancer cells(HeLa),and the ethyl ether extract of Jinsiyou showed stronger inhibitory ability against Staphylococcus aureus.It indicated that the chemical components of the eth-yl ether extracts from cultivated Aquilaria sinensis and wild Aquilaria sinensis were mainly sesquiter-penes and ketone compounds,but there were differences in the content and activity of characteristic components.
