通过动态监测北沙参种子休眠解除过程中的内源激素平衡变化、胚形态后熟进程与萌发特性演化三者间的时序关联与互作关系,系统揭示形态生理休眠的解除机制.以北沙参种子为材料进行质量检验,同时采用混沙层积与低温(5 ℃)裸层积两种方式处理种子,系统比较了其萌发指标、胚率及内源激素含量的动态变化.结果表明,这批北沙参种子的质量稳定且可靠,完全符合规模化生产所需的质量标准.层积处理能有效打破种子休眠,其中低温裸层积效果更佳,处理56 d即可使发芽率达100%,且显著促进胚率增长,完成形态后熟.生理机制上,抑制类激素脱落酸(ABA)含量持续下降,而促进激素(GA3、IAA、BR、CTK)含量及(GA3+IAA+BR+CTK+ETH)/ABA等比值显著上升,且裸层积处理能更快速地诱导激素平衡状态.说明5 ℃裸层积56 d是解除北沙参种子休眠的最佳方法,其本质是通过调控内源激素网络协同解除形态生理休眠.

By dynamically monitoring the changes in endogenous hormone balance during the process of seed dormancy breaking of Glehnia littoralis,the progress of embryo morphological after-ripening,and the evolution of germination characteristics,the temporal correlation and interaction relationships among these 3 aspects were systematically revealed,thus systematically elucidating the mechanism of seed dormancy breaking,which provided theoretical basis and technical support for the standardiza-tion and large-scale cultivation of Glehnia littoralis.The quality of Glehnia littoralis seeds was in-spected,and the seeds were treated with 2 methods:sand layering and low-temperature(5℃)bare layering.The dynamic changes in germination indicators,embryo rate,and endogenous hormone con-tents were systematically compared.The results showed that the quality of this batch of Glehnia litto-ralis seeds was stable and reliable,fully meeting the quality standards required for large-scale produc-tion.Layering treatment could effectively break seed dormancy.Among them,low-temperature bare layering had a better effect.After 56 days of treatment,the germination rate could reach 100%,and it significantly promoted the increase of embryo rate,completing the morphological after-ripening.From the physiological mechanism perspective,the content of inhibitory hormone ABA continuously decreased,while the contents of promoting hormones(GA3,IAA,BR,CTK)and the ratio of(GA3+IAA+BR+CTK+ETH)/ABA increased significantly,and bare layering treatment could in-duce the hormone balance state more quickly.It indicated that 5℃ bare layering for 56 days was the best method for breaking the dormancy of Glehnia littoralis seeds,and its essence was to regulate the endogenous hormone network and coordinate the release of morphological physiological dormancy.
