三黄三仙汤治疗再生障碍性贫血的作用机制研究OA
Study on the Mechanism of Sanhuang Sanxian Decoction(三黄三仙汤)in the Treatment of Aplastic Anaemia
目的:探讨三黄三仙汤治疗再生障碍性贫血(Aplastic Anemia,AA)的作用机制.方法:对脾淋巴细胞进行分离纯化,获取Naive CD4+T细胞,然后诱导产IL-17 的辅助性T细胞(IL-17-producing T helper,Th17)分化.采用流式细胞术分析CD4+T细胞的免疫表型,并通过光学显微镜评估其形态变化.CCK8 检测不同浓度三黄三仙汤含药血清处理后的细胞活力,流式细胞术鉴定各组调节性 T 细胞(Regulatory T cells,Treg)和Th17 细胞分化情况,采用酶联免疫吸附试验检测细胞中IL-17A、IL-10 和IL-21 的含量,qPCR检测细胞中IL-17F、T-bet mRNA表达,Western blot检测细胞中过氧化物酶体增殖物激活受体γ(Peroxisome proliferator-activated receptor γ,PPARγ)、类视黄醇相关孤儿受体γt(Retinoid-related orphan receptor gamma t,RORγt)、转录因子叉头盒蛋白3(Transcription factor forkhead box protein 3,Foxp3)的蛋白表达.结果:CCK8 结果显示,10%含药血清为最适血清浓度.与模型组比较,三黄三仙汤含药血清组、淫羊藿苷组中Th17 细胞占比显著下降,而Treg细胞的占比则显著上升;IL-17A和IL-21 的含量显著下降,IL-10 的含量显著上升;IL-17F、T-bet mRNA的表达水平显著降低;PPARγ、Foxp3蛋白表达显著升高,RORγt蛋白表达显著降低.而加入PPARγ抑制剂T0070907 后则出现相反的趋势.结论:三黄三仙汤能够有效改善AA,其作用机制可能与激活PPARγ,引起Foxp3/RORγt通路失调有关.
Objective:To explore the effect of Sanhuang Sanxian decoction in the treatment of aplastic anaemia(AA)and its underlying molecular mechanisms in depth.Methods:Splenic lymphocytes were isolated and purified to obtain Naive CD4+T cells,which were then induced to differentiate interleukin-17(IL-17)producing helper T cells(Th17).The immunophenotype of CD4+T cells was analyzed by flow cytometry,and their morphological changes were assessed by optical microscopy.The CCK8 assay detected cell viability after treatment with different concentrations of Sanhuang Sanxian decoction-containing serum.Flow cytometry was used to identify regulatory T cells(Treg)and Th17 cell differentiation in each group.An enzyme-linked immunosorbent assay was used to detect the levels of interleukin(IL)-17A,IL-10 and IL-21 in cells.qPCR was used to detect IL-17F,T-bet mRNA expression in cells.Western blot was performed to detect the protein expression of peroxisome proliferator-activated receptor γ(PPARγ),retinoid-related orphan receptor gamma t(RORγt),and transcription factor forkhead box protein 3(Foxp3)in cells.Results:CCK8 results indicated 10%drug-containing serum as the optimal serum concentration.Compared with the model group,there was a remarkable decreased proportion of Th17 cells in the Sanhuang Sanxian decoction-containing serum group and the Icariin group,whereas there was a significantly increase of the proportion of Treg cells.IL-17A and IL-21 content dramatically decreased and IL-10 content dramatically increased.The expression levels of IL-17F,T-bet mRNA was significantly reduced.The PPARγ and Foxp3 protein expression was markedly elevated,and the RORγt protein expression was markedly reduced.The opposite trend was observed with the addition of the PPARγ inhibitor T0070907.Conclusion:Sanhuang Sanxian decoction was able to effectively alleviated AA,and its mechanism of action may be related to the activation of PPARγ,causing dysregulation of the Foxp3/RORγt pathway.
