为了解青海省门源地区高原鼠兔(Ochotona curzoniae)体内人兽共患病原微生物的携带情况,并加深对青藏高原高海拔生态系统中高原鼠兔潜在病原携带特征的认识.本试验在青藏高原北部的门源县捕获60只高原鼠兔,分别采集其脾脏、肺脏和肠道内容物提取DNA,采用聚合酶链式反应(PCR)结合一代测序技术对多种病原微生物进行分子检测,并进行基因序列比对和系统发育分析.结果显示,布鲁氏菌检出率为3.3%(2/60),隐球菌为10.0%(6/60),毛霉菌为3.3%(2/60),艾美耳球虫为65.0%(39/60),囊孢球虫为10.0%(6/60),而耶尔森菌和弓形虫均未检出(0/60).进一步鉴定结果显示,布鲁氏菌为根际布鲁氏菌,隐球菌为大隐球菌和浅白隐球菌,毛霉菌为总状毛霉,囊孢球虫为赖氏囊孢球虫,而艾美耳球虫表现出较高的遗传多样性,包括3个独立的进化支和1个与Eimeria banffensis同源的进化支.结果表明,高原鼠兔体内可携带多种人兽共患病原体,具有潜在的传播风险,可能对人类健康构成威胁.本试验结果可为西北地区人兽共患疾病的传播机制研究和防控策略制定提供重要的科学依据.

To understand the prevalence of zoonotic pathogens in plateau pikas(Ochotona curzoniae)in Menyuan County,Qinghai Province,and to gain insights into the characteristics of pathogen carriage in high-altitude ecosystems of the Qinghai-Tibet Plateau,a molecular investigation was conducted.A total of 60 plateau pikas were captured in northern Qinghai-Tibet Plateau(Menyuan County).DNA was extracted from spleen,lung,and intestinal content samples,and polymerase chain reaction(PCR)combined with Sanger sequencing was used for molecular detection of multiple pathogens.Subsequent sequence alignment and phylogenetic analysis were performed.The results showed that the detection rates were:Brucella 3.3%(2/60),Cryptococcus 10.0%(6/60),Mucor 3.3%(2/60),Eimeria 65.0%(39/60),and Cystoisospora 10.0%(6/60),while Yersinia and Toxoplasma were not detected(0/60).Further species identification revealed that the Brucella species was Brucella rhizomaticola;the Cryptococcus species included C.neoformans and C.laurentii;the Mucor species was Rhizomucor pusillus;the Cystoisospora species was C.suis;and Eimeria showed high genetic diversity,with three independent evolutionary clades and one clade homologous to Eimeria banffensis.These findings indicate that plateau pikas can harbor multiple zoonotic pathogens and may pose a transmission risk to humans,potentially threatening public health.This study provides valuable scientific evidence for understanding the transmission mechanisms of zoonotic diseases and for the development of control strategies in Northwest China.
