新疆和云南地区节肢动物的RNA病毒组学分析OA
Metaviromic Analysis of RNA Viruses in Arthropods from Xinjiang and Yunnan
虫媒病毒经节肢动物传播,对人类健康和畜牧业生产构成威胁.为了解新疆与云南地区独特的地理和气候条件下虫媒病毒的多样性,并对其进行监测.本试验采用高通量测序(HTS)结合生物信息学方法,对新疆维吾尔自治区精河县和温泉县养殖场的蜱虫样本,以及云南省大理白族自治州养殖场的蚊子、苍蝇、白蛉样本开展宏病毒组学分析.结果显示,在204个节肢动物样本中,共鉴定到26个病毒科/目,35条完整或近乎完整病毒Contigs,包含8个脊椎动物相关病毒科,其中有5条Contigs与非冗余蛋白序列(NR)数据库中所有已注释病毒的病毒RNA依赖的RNA聚合酶(RdRp)编码区氨基酸序列的最高同源性均低于90%.云南省大理白族自治州蚊子样本的病毒多样性和丰度最高,以弹状病毒科(Rhabdoviridae)和中套病毒科(Mesoniviridae)为主,新疆维吾尔自治区的蜱虫样本中则以布尼亚病毒目(Bunyavirales)和楚病毒科(Chuviridae)为主.云南省蚊子样本中检出的南定(Nam dinh)病毒与越南脑炎患者以及深圳市和云南省检出的该病毒同源性极高(大于97%或99%).系统发育分析结果显示,部分新发现病毒与未分类病毒形成独立分支,可能为潜在新病毒属.结果表明,新疆和云南地区养殖场节肢动物所携带的病毒不仅具有丰富的遗传多样性,还存在潜在新病毒属和跨宿主传播风险.本试验结果可为保障畜牧业生产安全和公共卫生安全提供科学依据.
Arthropod-borne viruses are transmitted through arthropods and pose threats to both human health and livestock production.To investigate and monitor the diversity of arboviruses under the unique geographical and climatic conditions of Xinjiang and Yunnan,this study applied high-throughput sequencing(HTS)combined with bioinformatics to perform metaviromic analysis of tick samples collected from farms in Jinghe and Wenquan Counties,Xinjiang Uygur Autonomous Region,and mosquito,fly,and sandfly samples collected from farms in Dali Bai Autonomous Prefecture,Yunnan Province.A total of 204 arthropod samples yielded 26 viral families/orders and 35 complete or nearly complete viral Contigs,including 8 vertebrate-associated viral families.Among these,5 Contigs shared less than 90%amino acid sequence identity in the RNA dependent RNA polymerase(RdRp)-encoding region with any annotated viral sequences in the non-redundant protein sequences(NR)database.Viral diversity and abundance were highest in mosquito samples from Yunnan,dominated by the families Rhabdoviridae and Mesoniviridae,while tick samples from Xinjiang were dominated by Bunyavirales and Chuviridae.Notably,Nam dinh virus detected in Yunnan mosquito samples showed high homology(>97%or>99%)with strains previously reported in encephalitis patients in Vietnam,as well as strains detected in Shenzhen and Yunnan.Phylogenetic analysis revealed that some of the newly identified viruses clustered into independent branches with unclassified viruses,suggesting potential novel viral genera.These results demonstrate that arthropods from farms in Xinjiang and Yunnan harbor viruses with rich genetic diversity,including potential new genera and risks of cross-host transmission.This study provides a scientific basis for safeguarding livestock production and public health security.
