Identification and Drug Resistance Analysis of Clostridium perfringens Type A Isolated from Goat

为了查明河南省南阳市宛城区某羊场病死山羊的致病菌及其病原特性,本试验采集病死山羊肝脏、肺脏等病料,分离培养细菌,通过16S rDNA基因测序和遗传进化分析鉴定分离菌株的种属,通过PCR检测毒素基因确定其分型,对分离菌株进行多位点序列分型(MLST)、全基因组测序和遗传进化分析、单核苷酸多态性(SNP)分析,根据毒力因子数据库(VFDB)和综合抗生素耐药基因数据库(CARD)预测分离菌株的毒力基因和耐药基因;运用K-B琼脂纸片法检测分离菌株对12种抗菌药物的敏感性,并通过小鼠感染试验分析其致病性.结果显示,分离鉴定得到1株A型产气荚膜梭菌(命名为WC202409).全基因组遗传进化分析结果显示,该菌株与俄罗斯源产气荚膜梭菌(C.perfringens)QDAF01亲缘关系较近.SNP分析结果显示,分离菌株与我国菌株1718G、QHY-2、XZ-D1、ZWCP079的SNP差异数很小.VFDB注释到11种毒力因子和16种与产气荚膜梭菌相关的基因;CARD注释到6类抗生素14种耐药基因.药敏试验结果显示,分离菌株WC202409对四环素、头孢唑林、苯唑西林、青霉素G、链霉素和氟苯尼考耐药,对头孢噻肟、恩诺沙星、头孢曲松和亚胺培南敏感.小鼠致病性试验结果显示,试验组小鼠感染分离菌株5 d后全部死亡,剖检可观察到明显病变,经验证,致病菌为WC202409.结果表明,WC202409分离菌株为该羊群致病菌,对多种药物耐药,致病力强,可能来源于常年跨境贩运传播.本试验结果有助于产气荚膜梭菌病防控以及病原体和商业疫苗研究.

To identify the pathogen and its characteristics responsible for the death of goats in a sheep farm in Wancheng District,Nanyang City,Henan Province,liver and lung samples from diseased goats were collected for bacterial isolation and culture.The isolated strains were identified by 16S rDNA gene sequencing and phylogenetic analysis.Toxin genes were detected by PCR for typing.Multilocus sequence typing(MLST),whole-genome sequencing,phylogenetic and SNP analysis were performed.Virulence genes and antimicrobial resistance genes were predicted using the VFDB and CARD databases.The Kirby-Bauer(K-B)disk diffusion method was used to test the strain's susceptibility to 12 antimicrobial agents,and a mouse infection test was conducted to evaluate pathogenicity.One strain of Clostridium perfringens type A was isolated and named WC202409.Phylogenetic analysis revealed that the strain is closely related to the Russian strain QDAF01.SNP analysis showed minimal variation between WC202409 and Chinese strains 1718G,QHY-2,XZ-D1,and ZWCP079.VFDB analysis annotated 11 virulence factors and 16 C.perfringens-related genes,while CARD predicted 14 resistance genes associated with six classes of antibiotics.Antimicrobial susceptibility testing showed resistance to tetracycline,cefazolin,oxacillin,penicillin G,streptomycin,and florfenicol,but susceptibility to cefotaxime,enrofloxacin,ceftriaxone,and imipenem.In the mouse infection test,all mice died within 5 days,with obvious pathological lesions confirmed to be caused by WC202409.The results indicate that strain WC202409 was the causative agent of disease in the affected goat flock,exhibits multidrug resistance,and is highly pathogenic.The strain may have originated from transboundary livestock trade.These findings provide valuable reference for the prevention and control of C.perfringens infection and for vaccine development and pathogen research.
