低聚糖对蛋鸡生产性能、抗氧化和免疫功能的影响OA
Effects of oligosaccharides on the production performance,antioxidant,and immunefunction of laying hens
旨在评价不同低聚糖对蛋鸡生产性能、抗氧化和免疫功能的影响.选用 448 只 500 日龄健康海兰灰蛋鸡,随机分为 4 组,每组设 8 个重复,每个重复 14 只,分别饲喂基础日粮(CON)和基础日粮中分别添加20 mg/kg低聚壳寡糖(COS)、50 mg/kg低聚木糖(XOS)及 500 mg/kg甘露低聚糖(MOS)的试验日粮,试验期为 56 d.结果:与CON组相比,COS、XOS和MOS组蛋鸡平均日采食量均显著降低(P<0.05),蛋鸡血清中白蛋白(ALB)含量与总抗氧化能力(T-AOC)水平显著提高(P<0.05),而低密度脂蛋白胆固醇含量显著降低(P<0.05);COS提高了血清总超氧化物歧化酶(T-SOD)活性和肝脏过氧化氢酶(CAT)活性(P<0.05),MOS 提高了卵巢T-SOD活性和子宫T-AOC 水平(P<0.05);COS和MOS提高了蛋鸡肝脏和卵巢核因子E2 相关因子 2(Nrf2)的mRNA表达量(P<0.05);COS、XOS和MOS均提高了肝脏超氧化物歧化酶1(SOD1)的mRNA表达量(P<0.05),而MOS提高了卵巢SOD1 的mRNA表达量(P<0.05);3 种低聚糖使肝脏谷胱甘肽过氧化物酶1(GPX1)的mRNA表达量均显著升高(P<0.05);COS和XOS使肝脏过氧化氢酶(CAT)的mRNA表达量显著升高(P<0.05),COS使卵巢CAT的mRNA表达量显著升高(P<0.05),而XOS使卵巢CAT的mRNA表达量显著降低(P<0.05);COS和MOS使肝脏和卵巢血红素加氧酶-1(HO-1)的mRNA表达量显著升高(P<0.05);XOS和MOS提高了血清免疫球蛋白A(IgA)含量(P<0.05),COS提高了血清免疫球蛋白G(IgG)含量(P<0.05),3 种低聚糖均降低了血清白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量(P<0.05).综上,日粮中添加COS、XOS和MOS均能增强蛋鸡抗氧化功能,调节肝脏和卵巢相关基因表达,提高机体免疫力,改善蛋鸡健康.
This study aimed to evaluate the effects of different oligosaccharides on the production performance,antioxidant capacity,and immune function of laying hens.A total of 448 healthy 500-day-old laying hens were obtained and randomly assigned to 4 groups,with 8 replicates per group and 14 hens per replicate.The hens were fed with either a basal diet(CON)or experimental diets supplemented with 20 mg/kg chitosan oligosaccharides(COS),50 mg/kg xylooligosaccharides(XOS),or 500 mg/kg mannan oligosaccharides(MOS).The ex-perimental period lasted for 56 days.The results showed that,compared with the CON group,the COS,XOS,and MOS groups significantly reduced the average daily feed intake of the laying hens(P<0.05).These oligosaccharides significantly increased the serum albumin content and total antioxidant capacity levels(T-AOC)in the birds,but reduced the serum low-density lipoprotein cholesterol content in them(P<0.05).Specifically,COS enhanced serum total superoxide dismutase(T-SOD)and hepatic catalase(CAT)activities(P<0.05),whereas MOS improved ovarian T-SOD activity and uterine T-AOC levels(P<0.05)of the hens.COS and MOS up-regulated the mRNA expression of nuclear factor erythroid 2-related factor 2(Nrf2)in both the liver and ovaries(P<0.05)of the hens.All three oligosaccharides up-regu-lated hepatic superoxide dismutase 1(SOD1)mRNA expression(P<0.05),and MOS also elevated its expression in their ovaries.The mRNA expression of hepatic glutathione peroxidase 1(GPX1)in the hens was significantly enhanced by all supplements(P<0.05).For cat-alase(CAT),COS and XOS up-regulated its hepatic mRNA expression,while COS up-regulated and XOS down-regulated its ovarian ex-pression(P<0.05).Both COS and MOS significantly increased the mRNA expression of heme oxygenase-1(HO-1)in the liver and ovaries(P<0.05).XOS and MOS increased serum immunoglobulin A(IgA)content,and COS increased immunoglobulin G(IgG)content(P<0.05).All the three oligosaccharides significantly reduced the serum content of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)(P<0.05).In conclusion,dietary supplementation with COS,XOS,and MOS improved antioxidant status,regulated gene ex-pression in the liver and ovary,and enhanced immune parameters and overall health of the laying hens.
