宣和猪ZFAT基因外显子7多态性及其对胴体和肉质性状的影响OA
Polymorphisms of the exon 7 of ZFAT gene and its effects on carcass and meat traits in Xuanhe pigs
旨在探讨含AT-hook结构域的锌指蛋白(ZFAT)基因外显子 7 多态性及其对宣和猪胴体和肉质性状的影响.利用一代测序技术检测了106 头宣和猪ZFAT基因外显子7 的单核苷酸多态性(SNP)位点,通过最小二乘模型分析各SNP 位点的基因型及单倍型组合与胴体和肉质性状的关联.结果:在ZFAT基因外显子7 区域共检测出1 个错义突变(G3174A)和4 个同义突变(C2693T、C2834T、T3173C和C3224T)位点,且都处于哈迪-温伯格平衡;除C2693T和G3174A位点外,C2834T、T3173C和C3224T位点均表现为中度多态(0.307 6≤多态信息含量(PIC)≤0.373 9),遗传多样性丰富(0.379 7≤杂合度(H)≤0.497 8);关联分析结果显示,C2693T、C2834T、T3173C位点与平均背膘厚和后腿比例间存在显著关联(P<0.05 或P<0.01),C2693T、C2834T、T3173C和G3174A位点与肉色评分、pH值、滴水损失、肌内脂肪含量间存在显著关联(P<0.05 或P<0.01);不同单倍型组合与胴体直长、胴体斜长、后腿比例、瘦肉率、屠宰率等胴体性状,以及肉色评分、大理石纹评分、pH值、熟肉率、滴水损失等肉质性状存在显著差异(P<0.05 或P<0.01),CCCGT和CCTGC单倍型分别对胴体和肉质水平表现出明显优势.研究结果初步揭示了ZFAT基因外显子7 多态性与宣和猪的胴体和肉质性状间存在显著关联,为利用该基因标记改良猪胴体和肉质性状提供了理论依据.
To identify the polymorphisms of exon 7 in the ZFAT gene and its effect on meat traits in Xuanhe pigs,this study used first se-quencing technology to detect single nucleotide polymorphisms(SNPs)in exon 7 of the ZFAT gene in 106 Xuanhe pigs.Different genotypes and haplotype combinations were then associated with carcass and meat quality traits using a least squares model.The results showed that one missense mutation site(G3174A)and four synonymous mutation sites(C2693T,C2834T,T3173C,and C3224T)were identified in the ex-on 7 region of the ZFAT gene,and all were in a Hardy-Weinberg equilibrium state(P>0.05).Except the C2693T and G3174A sites,the C2834T,T3173C and C3224T sites exhibited moderate polymorphism(0.3076≤PIC≤0.3739)and rich genetic diversity(0.3797≤H≤0.497 8)in Xuanhe pigs.The results of the association analysis showed that the C2693T,C2834T and T3173C sites were significantly asso-ciated with average backfat thickness and ham percentage(P<0.05 or P<0.01)in Xuanhe pigs.The C2693T,C2834T,T3173C and G3174A sites were significantly associated with meat color score,pH value,drip loss,and intramuscular fat content(P<0.05 or P<0.01)in Xuanhe pigs.Different haplotype combinations were significantly associated with carcass traits(carcass straight length,carcass diagonal length,ham percentage,lean percentage,dressing percentage),and meat traits(meat color score,marbling score,pH value,cooked rate,and drip loss)(P<0.05 or P<0.01)in the pigs.Meanwhile,the individuals carrying CCCGT and CCTGC haplotypes exhibited high carcass and meat quality.These results indicated that polymorphisms of exon 7 in ZFAT gene were significantly associated with carcass and meat traits in Xuanhe pigs,which would serve as a theoretical basis for using this gene marker to improve the meat quality traits in pigs.
