PPE61是结核分枝杆菌特有的PPE蛋白家族成员,它的可变C端与分泌特性暗示其在免疫逃逸和宿主互作中起关键作用.然而,该蛋白对呼吸道屏障上皮细胞的具体功能未知.本研究通过体外试验,旨在阐明PPE61对人支气管上皮细胞(BEAS-2B)增殖的调控作用及其在感染早期的分子机制,为结核病诊断与疫苗研发提供理论依据.采用生物信息学软件预测PPE61蛋白的基本生物学性质,基于NCBI数据库获取结核分枝杆菌PPE61基因及其编码蛋白序列信息.通过PCR从卡介苗(BCG)基因组中扩增出PPE61基因,并构建了重组表达质粒pET22b(+)-PPE61,并转化至大肠杆菌BL21(DE3)宿主中进行蛋白表达.通过SDS-PAGE和Western blot验证蛋白表达及抗原性;采用CCK-8法检测不同质量浓度(2.5~20 μg/mL)PPE61处理BEAS-2B细胞24h的增殖活性,应用qPCR分析TGF-β基因表达水平.结果显示:PPE61基因扩增后全长1 287 bp,编码406个氨基酸,该蛋白无信号肽和跨膜区,预测含37个磷酸化位点,41个B细胞抗原表位和35个T细胞抗原表位,二级结构以α-螺旋(49.26%)为主;经SDS-PAGE分析发现该蛋白以包涵体形式存在,相对分子质量为105.652 18×105,与预期大小相符合,说明重组质粒转化大肠杆菌BL21(DE3)感受态细胞后,能诱导表达目标蛋白.Western blot证实PPE61抗原具有特异性;其浓度依赖性抑制细胞增殖(P≤0.000 1),可显著上调TGF-β表达(P<0.05).结果表明:PPE61蛋白具备丰富的抗原表位和高可溶性表达特性,预示其拥有良好的免疫原性与开发潜力,为筛选结核病诊断标志物和研发新型亚单位疫苗提供了新靶点,奠定了重要的试验基础.

PPE61,a member of the PPE protein family specific to Mycobacterium tuberculosis,plays a key role in immune evasion and host-pathogen interactions,as implicated by its variable C-terminal region and secretion properties.However,its specific functions in respiratory barrier epithelial cells remains unclear.In order to explore,through in vitro experiments,the regulatory role of PPE61 on the proliferation of human bronchial epithelial cells(BEAS-2B)and its molecular mechanisms during early infection,and to provide a theoretical basis for tuberculosis diagnosis and vaccine development,the fundamental biological properties of PPE61 protein were predicted using bioinformatics software,PPE61 gene and its encoded protein sequence information were obtained from NCBI database.PPE61 gene was amplified via PCR from Bacillus Calmette-Guérin(BCG)genome,and a recombinant expression plasmid,pET-22b(+)-PPE61,was constructed and transformed into Escherichia coli(E.col)BL21(DE3)for protein expression.Protein expression and antigenicity were verified by SDS-PAGE and Western blot;the proliferative activity of BEAS-2B cells treated with different concentrations(2.5-20 μg/mL)of PPE61 for 24 hours was detected using CCK-8 assay,and the level of TGF-β gene expression was analyzed by qPCR.The results showed that the amplified PPE61 gene was 1 287 bp in length,encoding 406 amino acids,the protein lacked a signal peptide and transmembrane regions,but was predicted to contain 37 phosphorylation sites,41 B-cell epitopes and 35 T-cell epitopes,and its secondary structure was predominantly alpha-helical(49.26%);SDS-PAGE analysis revealed that the protein was available in the form of inclusion body with an approximate molecular weight of 105.652 18×105,consistent with its expected size,indicating that target protein could be induced and expressed after the recombinant plasmid was transformed into competent E.coli BL21(DE3)cell.It was confirmed that,by Western blot,PPE61 antigen was specific;its dose-dependently inhibited cell proliferation(P≤0.000 1)and significantly upregulated TGF-βexpression(P<0.05).In conclusion,PPE61 protein had rich antigenic epitopes and high solubility,indicating good immunogenicity and promising prospects for development,by which a new target was provided for screening markers of tuberculosis diagnosis and developing novel subunit vaccines,and an important experimental foundation was laid.
