目的 揭示芝麻林素(Ses)对糖尿病视网膜病变(DR)大鼠的治疗作用及其对p38丝裂原活化蛋白激酶(MAPK)信号通路介导炎症反应的影响.方法 将SPF级雄性SD大鼠腹腔注射35 mg·kg-1链脲佐菌素构建DR模型大鼠.大鼠随机分为6组:对照组、DR 组、10/20/40-Ses 组(分别灌胃 10、20、40 mg·kg-1 的 Ses)和 40-Ses+Anisomycin 组(灌胃40 mg·kg-1的Ses,同时尾静脉注射2 mg·kg-1 p38 MAPK激动剂Anisomycin),每组12只.对照组为正常饲养大鼠,其他组均为DR模型大鼠.干预周期为4周.使用血糖仪检测大鼠空腹血糖(FBG);使用酶联免疫吸附测定(ELISA)试剂盒检测大鼠糖化血红蛋白(HbA1c)和空腹胰岛素(FINS)表达水平;对大鼠视网膜进行苏木素伊红(HE)和TUNEL染色用于观察其视网膜形态和细胞凋亡;使用ELISA法和qRT-PCR检测大鼠视网膜炎症相关指标[肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、IL-6、细胞间黏附分子-1(ICAM-1)以及单核细胞趋化蛋白-1(MCP-1)]蛋白与mRNA表达水平;使用qRT-PCR检测大鼠视网膜Bax和Bcl-2 mRNA表达水平;使用Western blot检测大鼠视网膜p38 MAPK和p65核因子-KB(p65 NF-κB)磷酸化表达水平.结果 对照组大鼠视网膜结构组织清晰完整,DR组大鼠视网膜结构排列紊乱松散,10/20/40-Ses组大鼠视网膜结构排列有所改善,40-Ses+Anisomycin组大鼠视网膜病变加重.与对照组比较,DR组大鼠视网膜FPG、HbA1c、FINS、Bax mRNA 表达水平与 TUNEL+细胞比例均升高,TNF-α、IL-1β、IL-6、ICAM-1以及MCP-1蛋白和mRNA表达水平以及p38 MAPK与p65 NF-κB磷酸化水平均升高,Bcl-2 mRNA相对表达水平降低,差异均有统计学意义(均为P<0.05).与DR组比较,10/20/40-Ses组大鼠视网膜FPG、HbA1c、FINS、Bax mRNA表达水平与TUNEL+细胞比例均降低,TNF-α、IL-1β、IL-6、ICAM-1、MCP-1蛋白和mRNA表达水平以及p38 MAPK与p65 NF-κB磷酸化水平均降低,Bcl-2 mRNA相对表达水平均升高,差异均有统计学意义(均为P<0.05).与40-Ses组比较,40-Ses+Anisomycin组大鼠视网膜FPG、HbA1c、FINS、Bax mRNA表达水平与TUNEL+细胞比例均升高,TNF-α、IL-1β、IL-6、ICAM-1、MCP-1蛋白和mRNA表达水平以及p38 MAPK与p65 NF-κB磷酸化水平均升高,Bcl-2 mRNA相对表达水平降低,差异均有统计学意义(均为P<0.05).结论 Ses通过抑制DR中p38 MAPK信号通路的激活减轻过度炎症反应,从而发挥治疗作用.

Objective To reveal the therapeutic effect of sesamolin(Ses)on diabetic retinopathy(DR)in rats and its effect on the inflammatory response mediated by p38 mitogen-activated protein kinase(MAPK)signaling pathway.Methods SPF-grade male SD rats were intraperitoneally injected with 35 mg·kg-1 streptozotocin to establish DR model rats.Rats were randomly divided into six groups:control group,DR group,10/20/40-Ses group(gavage of 10,20,and 40 mg·kg-1 Ses,respectively),and 40-Ses+Anisomycin group(gavage of 40 mg·kg-1Ses,and tail vein injection of 2 mg·kg-1 p38 MAPK agonist Anisomycin),with 12 rats in each group.The control group consisted of normally fed rats,while the other groups consisted of DR model rats.The intervention lasted 4 weeks.The fasting blood glucose(FBG)in rats was measured using a blood glucose meter;the expression levels of glycated hemoglobin(HbA1c)and fasting insulin(FINS)in rats were detected using the enzyme-linked immunosorbent assay(ELISA)kit.Hematoxylin-eosin and TUNEL staining were per-formed on the retina of rats to observe its morphology and cell apoptosis.ELISA and quantitative real-time polymerase chain reaction(qRT-PCR)were employed to detect the protein and mRNA expression levels of inflammation-related indicators in rat retina,including tumor necrosis factor-α(TNF-α),interleukin(IL)-1β,IL-6,intercellular adhesion molecule-1(ICAM-1),and monocyte chemoattractant protein-1(MCP-1);qRT-PCR was used to detect the mRNA expression levels of Bax and Bcl-2 in rat retina;Western blot was applied to detect the expression levels of phosphorylated p38 MAPK and p65 nuclear factor-κB(p65 NF-NF)in rat retina.Results The retinal structure of rats in the control group was clear and intact,while the retinal structure of rats in the DR group was disordered and loose.The retinal structure of rats in the 10/20/40-Ses group improved,while the retinal lesions of rats in the 40-Ses+Anisomycin group worsened.Compared with the control group,the FPG,HbA1c,FINS,Bax mRNA expression levels,and the TUNEL+cell ratio in the rat retina in the DR group increased;the expression levels of TNF-α,IL-1β,IL-6,ICAM-1,and MCP-1 protein and mRNA,as well as the phosphorylation levels of p38 MAPK and p65 NF-κB,increased;the relative expression level of Bcl-2 mRNA decreased,and the differences were all statisti-cally significant(all P<0.05).Compared with the DR group,the FPG,HbA1c,FINS,Bax mRNA expression levels,and the TUNEL+cell ratio in the retina of rats in the 10/20/40-Ses group decreased;the expression levels of TNF-α,IL-1β,IL-6,ICAM-1,and MCP-1 protein and mRNA,as well as the phosphorylation levels of p38 MAPK and p65 NF-κB,decreased;the relative expression level of Bcl-2 mRNA increased,and the differences were all statistically significant(all P<0.05).Com-pared with the 40-Ses group,the FPG,HbA1c,FINS,Bax mRNA expression levels,and the TUNEL+cell ratio in the retina of rats in the 40-Ses+Anisomycin group increased;the expression levels of TNF-α,IL-1β,IL-6,ICAM-1,and MCP-1 protein and mRNA,as well as the phosphorylation levels of p38 MAPK and p65 NF-κB,increased;the relative expression level of Bcl-2 mRNA decreased,and the differences were all statistically significant(all P<0.05).Conclusion Ses exerts thera-peutic effects by inhibiting the activation of the p38 MAPK signaling pathway in DR and reducing excessive inflammatory re-sponses.
