目的 探讨滋阴明目方(ZY)对干性年龄相关性黄斑变性(dAMD)模型小鼠的保护作用及机制.方法 采用高脂饮食联合碘酸钠注射建立小鼠dAMD模型.将60只小鼠随机分为6组:空白组、模型组、ZY-低剂量组(ZY-L组)、ZY-中剂量组(ZY-M组)、ZY-高剂量组(ZY-H组)以及Lutein组,每组10只.空白组普通饲料喂养3个月后腹腔注射生理盐水,其余各组用高脂饲料喂养3个月后,腹腔注射碘酸钠(20 mg·kg-1).ZY低、中、高剂量组每日分别给予ZY中药液(剂量分别为11、22、44 g·kg-1)灌胃1次,连续干预2周,空白组与模型组小鼠予生理盐水灌胃处理.使用光学相干断层扫描、眼底自发荧光以及HE染色分析评估小鼠视网膜损伤;使用转录组学分析小鼠眼球组织差异表达基因;免疫荧光染色检测小鼠眼球组织中磷酸化核糖体蛋白S6激酶1(p-p70S6K)表达水平;ELISA测定小鼠血清中衰老相关分泌表型关键炎症因子相关蛋白[白细胞介素-6(IL-6)、单核细胞趋化蛋白1(MCP-1)以及基质金属蛋白酶-9(MMP-9)]表达水平;Western blot检测小鼠视网膜mTOR通路相关蛋白[磷酸化哺乳动物雷帕霉素靶蛋白(p-mTOR)、p-p70S6K、肿瘤蛋白53(P53)以及细胞周期蛋白依赖性激酶抑制剂1 A(P21)]表达水平.结果 影像学检测结果显示,ZY干预显著改善了 dAMD小鼠模型的视网膜损伤,缩小了萎缩区域,减少了玻璃膜疣样沉积,改善了视网膜变薄.转录组学分析结果显示,与模型组相比,在ZY-H组中共1 592个差异表达基因,KEGG富集分析中,主要富集的通路包括甘油磷脂代谢通路、mTOR信号通路、谷胱甘肽代谢通路、脂肪细胞因子信号通路以及血管内皮生长因子信号通路等.免疫荧光染色检测结果显示,与模型组相比,ZY-H组与Lutein组p-p70S6K免疫荧光染色强度均下降,差异均有统计学意义(均为P<0.01).ELISA检测结果表明,与模型组比较,ZY-H组及Lutein组小鼠血清中IL-6、MMP-9以及MCP-1表达水平均下降,差异均有统计学意义(均为P<0.05).Western blot检测结果显示,与模型组比较,ZY-H组及Lutein组小鼠视网膜组织中p-mTOR及p-p70S6K表达水平均下降,ZY-M组、ZY-H组以及Lutein组小鼠视网膜组织中P53及P21表达水平均下降,差异均有统计学意义(均为P<0.05).结论 ZY通过抑制mTORC1/p70S6K通路,降低P53与P21等细胞衰老关键因子以及衰老相关分泌表型,对dAMD小鼠模型视网膜发挥保护作用.

Objective To explore the protective effect of Ziyin Mingmu Formula(ZY)on mice with dry age-related macular degeneration(dAMD)and its underlying mechanism.Methods A mouse dAMD model was established using a high-fat diet combined with sodium iodate injection.Sixty mice were randomly divided into six groups:blank group,model group,ZY-low-dose group(ZY-L group),ZY-medium dose group(ZY-M group),ZY-high-dose group(ZY-H group),and Lu-tein group,with 10 mice in each group.Mice in the blank group were fed with regular feed for 3 months and then intraperito-neally injected with physiological saline,while mice in the other groups were fed with high-fat feed for 3 months and then intraperitoneally injected with sodium iodate(20 mg·kg-1).Mice in the ZY-L,ZY-M,and ZY-H groups were given ZY tradi-tional Chinese medicine solution at doses of 11,22,and 44 g·kg-1,respectively,by gavage once a day for 2 consecutive weeks,while mice in the blank group and the model group were treated with physiological saline by gavage.The retinal inju-ry of mice was evaluated using optical coherence tomography,fundus autofluorescence,and hematoxylin-eosin staining;dif-ferentially expressed genes in mouse eyeball tissue were analyzed using transcriptomics;the expression level of phosphoryl-ated ribosomal protein S6 kinase 1 in mouse eyeball tissue was detected using immunofluorescence staining;the expression levels of key inflammatory factor related proteins[interleukin-6(IL-6),monocyte chemoattractant protein-1(MCP-1),and matrix metalloproteinase-9(MMP-9)]in the serum of mice with age-related secretory phenotypes were measured using en-zyme-linked immunosorbent assay(ELISA);the expression levels of mTOR pathway related proteins in mouse retina,inclu-ding phosphorylated mammalian target of rapamycin(p-mTOR),p-p70S6K,tumor protein 53(P53),and cyclin dependent kinase inhibitor 1A(P21),were determined using Western blot.Results The imaging results showed that ZY intervention significantly alleviated retinal damage in the dAMD mouse model,reduced atrophy area,decreased verrucous deposition on the vitreous membrane,and improved retinal thinning.The transcriptomic analysis results showed that compared with the model group,there were 1 592 differentially expressed genes in the ZY-H group,and according to the KEGG enrichment analysis,they were mainly enriched in glycerophospholipid metabolism pathway,mTOR signaling pathway,glutathione metabolism pathway,adipocytokine signaling pathway,and vascular endothelial growth factor signaling pathway.The immu-nofluorescence staining results showed that compared with the model group,the immunofluorescence staining intensity of p-p70S6K decreased in both the ZY-H group and the Lutein group,and the differences were statistically significant(both P<0.01).The ELISA results showed that compared with the model group,the expression levels of IL-6,MMP-9,and MCP-1 in the serum of mice in the ZY-H and Lutein groups decreased,and the differences were statistically significant(all P<0.05).The Western blot results showed that compared with the model group,the expression levels of p-mTOR and p-p70S6K in the retinal tissue of mice in the ZY-H and Lutein groups decreased;the expression levels of P53 and P21 in the retinal tissue of mice in the ZY-M,ZY-H,and Lutein groups decreased,and the differences were statistically significant(all P<0.05).Con-clusion ZY exerts a protective effect on the retina of the dAMD mouse model by inhibiting the mTORC1/p-70S6K path-way and reducing cell aging-related key factors such as P53 and P21,as well as aging-related secretory phenotypes.
