芦丁对氧化损伤环境下晶状体透明度的影响及其作用机制OA

目的 探究芦丁对氧化损伤环境下大鼠晶状体体外培养模型透明度的影响及其作用机制.方法 取60只8周龄SD大鼠双眼的完整晶状体,排除外伤致混浊的晶状体后,取10枚晶状体用50 μmol·L-1芦丁干预为芦丁组,仅用来观察晶状体透明度变化,余晶状体随机分为3组进行后续实验,分别为对照组、过氧化氢(H2O2)组和H2O2+芦丁组.暗场显微镜下观察晶状体的混浊情况,ImageJ软件对各组晶状体的混浊度进行统计分析.Western blot实验检测各组大鼠晶状体内质网应激蛋白PERK、IRE1α和ATF6的表达;免疫荧光染色观察各组大鼠晶状体活性氧(ROS)的含量;钙离子和脂质氧化损伤检测试剂盒检测各组大鼠晶状体钙离子和丙二醛(MDA)的含量.结果 显微镜下观察发现,芦丁组大鼠晶状体混浊时间与对照组(7 d)相比延缓至9 d.体外培养5 d时,与对照组大鼠晶状体混浊度(1.03±0.07)相比,H2O2组大鼠晶状体混浊度(6.89±0.15)显著增加(P<0.001);与H2O2组相比,H2O2+芦丁组大鼠晶状体混浊度(2.52±0.15)显著降低(P<0.001).免疫荧光检测发现,H2O2+芦丁组大鼠晶状体ROS相对荧光强度(4.35±0.11)较H2O2组(8.53±0.44)显著下降,差异有统计学意义(P<0.001).与H2O2组相比,H2O2+芦丁组晶状体内质网应激相关蛋白PERK、IRE1α、ATF6的相对表达量均显著降低,差异均有统计学意义(均为P<0.001).钙离子和脂质氧化损伤实验检测结果显示,H2O2+芦丁组大鼠晶状体钙离子含量(0.09±0.01)mmol·L-1和MDA含量(6.82±0.42)μmol·L-1 均较 H2O2 组钙离子含量(0.24±0.03)mmol·L-1 和 MDA 含量(10.63±1.13)μmol·L-1明显下降,差异均有统计学意义(均为P<0.01).结论 在H2O2诱导的氧化损伤环境下,芦丁通过抑制内质网应激来保护大鼠晶状体的内质网功能正常运行,从而延缓晶状体混浊的发生.

Objective To investigate the effect of rutin on the transparency of rat lens models cultured in vitro under oxidative stress and its underlying mechanism.Methods In this study,the intact binocular lenses of 60 8-week-old SD rats were taken.After excluding the lenses with turbidity caused by trauma,10 lenses were selected for intervention with 50μmol·L-1 rutin as the rutin group,which was only used to observe the changes in the transparency of the lenses.The re-maining lenses were randomly divided into 3 groups for subsequent experiments:the control group,the hydrogen peroxide(H2O2)group,and the H2O2+rutin group.The opacity of the lenses in each group was observed under a dark-field micro-scope and then statistically analyzed using ImageJ software.The expressions of endoplasmic reticulum stress proteins like PERK,IRE1α,and ATF6 in lenses were detected by Western blot.The content of reactive oxygen species(ROS)in lenses was measured using immunofluorescence staining,while the content of calcium ions and malondialdehyde(MDA)in lenses was measured using the calcium ion and lipid oxidative damage detection kit.Results Microscopic observations revealed that the lens opacity time of rats in the rutin group was delayed to 9 days compared with the control group(7 days).After 5 days of culture in vitro,the lens opacity of rats in the H2O2 group(6.89±0.15)significantly increased compared with that in the control group(1.03±0.07)(P<0.001);compared with the H2O2 group,the lens opacity of rats in the H2O2+rutin group(2.52±0.15)significantly decreased(P<0.001).Immunofluorescence staining showed that the relative fluorescence intensity of ROS in rat lenses in the H2O2+rutin group(4.35±0.11)significantly decreased compared to the H2O2 group(8.53±0.44),and the difference was statistically significant(P<0.001).Compared with the H2O2 group,the relative ex-pression levels of endoplasmic reticulum stress-related proteins PERK,IRE1a,and ATF6 in rat lenses in the H2O2+rutin group were significantly reduced,with all differences being statistically significant(all P<0.001).The results from the cal-cium ion and lipid oxidative damage assays showed that the calcium ion content(0.09±0.01)mmol·L-1 and MDA con-tent(6.82±0.42)μmol·L-1 in rat lenses in the H2O2+rutin group were remarkably lower than those in the H2O2 group,which had a calcium ion content of(0.24±0.03)mmol·L-1 and an MDA content of(10.63±1.13)μmol·L-1,and the differences were both statistically significant(both P<0.01).Conclusion Under the oxidative damage induced by H2O2,rutin protects the normal function of the endoplasmic reticulum in rat lenses by inhibiting endoplasmic reticulum stress,thereby delaying the occurrence of lens opacity.
