该文设计合成金属有机骨架化合物衍生的Co/NC纳米酶检测葡萄糖含量的综合实验,采用一系列仪器表征Co/NC纳米酶,通过控制变量法、动力学测试和标准曲线法优化纳米酶测试条件和检测葡萄糖范围.实验结果表明,当钴锌的物质的量比为 1:1,碳化温度为 900℃,pH值为 3.5时,Co/NC纳米酶的类氧化酶活性最大;Co/NC纳米酶最大反应速率为 1.74×10-7 mol·L-1·s-1,且对底物TMB有很强亲和力;检测葡萄糖的线性范围为 1~60 μmol,且检测体系具有优异的选择性.该实验包含了多学科知识点,有助于学生融会贯通知识体系,开拓科学研究思维,提升创新能力,培养科学素养.

In this paper,a comprehensive experiment is designed to synthesize Co/NC nanozymes derived from metal-organic frameworks for the detection of glucose content.A series of instruments are used to characterize the Co/NC nanozymes.Through control variable method,kinetic tests,and standard curve method,the testing conditions of nanozymes and the detection range for glucose are optimized.The experimental results show that when the molar ratio of Co to Zn is 1:1,the carbonization temperature is 900℃,and pH is 3.5,the Co/NC nanozymes exhibit the greatest oxidase-like activity.The maximum reaction rate of Co/NC nanozymes is 1.74×10-7 mol·L-1·s-1,and the nanozymes demonstrate a strong affinity for the substrate TMB.The linear range for glucose detection is 1~60 μmol,and the detection system exhibits excellent selectivity.The experiment integrates multidisciplinary knowledge,helping students achieve a comprehensive understanding of the knowledge system,develop scientific research thinking,improve innovation ability,and cultivate scientific literacy.
