咖啡酸对食管鳞癌细胞内质网应激及恶性增殖能力的影响OA
Effect of Caffeic Acid on Endoplasmic Reticulum Stress and Malignant Proliferative Capacity in Esophageal Squamous Cell Carcinoma Cells
目的 分析咖啡酸(CA)对食管鳞癌(ESCC)细胞恶性增殖能力的影响,并探索其潜在作用机制.方法 采用不同浓度(10、20 和 50 μM)CA处理ESCC细胞(KYSE140 及KYSE150),实验分为对照组及不同浓度CA观察组.通过试剂盒检测各组细胞中氧化应激指标(ROS、MDA及SOD);采用CCK8 及平板克隆检测各组细胞的体外增殖能力;利用Western Blot检测各组细胞中内质网应激标志蛋白GRP78 的表达情况.结果 与对照组相比,不同浓度(10、20 和 50 μM)CA观察组细胞的ROS及MDA含量逐渐降低、SOD活性逐渐增高、细胞的体外增殖能力逐渐减弱且GRP78 蛋白表达量逐渐减少(P<0.05).结论 CA可通过其抗氧化作用抑制ESCC细胞内质网应激,从而遏制这些细胞的恶性增殖.
Objective To analyze the effect of caffeic acid(CA)on the malignant proliferative capacity of esophageal squamous cell carcinoma(ESCC)cells and explore its underlying mechanism.Methods ESCC cells(KYSE140 and KYSE150)were treated with different concentrations of CA(10,20,and 50 μM).The experiment was divided into a control group and different CA observation groups.Oxidative stress markers(ROS,MDA,and SOD)in cells from each group were detected using assay kits.The in vitro proliferative capacity of cells in each group was assessed using CCK8 and plate colony formation assay.The expression of the endoplasmic reticulum stress marker protein GRP78 in each group was detected by Western Blot.Results Compared with the control group,cells in the CA observation groups(10,20,and 50 μM)showed a progressive decrease in ROS and MDA content,a gradual increase in SOD activity,a gradual weakening ofin vitro proliferative capacity,and a gradual decrease in GRP78 protein expression(P<0.05).Conclusion CA can inhibit endoplasmic reticulum stress in ESCC cells through its antioxidant effects,thereby suppressing the malignant proliferation of these cells.
