乌龟苗种低温保存技术研究OA
Research on cryopreservation of Mauremys reevesii hatchlings
为探究低温保存对乌龟稚龟生长的影响,采用人工控温方式,将稚龟置于10℃条件下分别保存0、30、60、90 d,统计各组存活率后,将其转移至29℃环境养殖10 d,测定生化指标和运动性能,并对低温处理后的稚龟在 29℃条件下养殖 2 个月并测定生长速率.结果显示,10℃低温保存 0、30、60、90 d组稚龟存活率依次为 100%、100%、97.78%、42.22%,低温处理结束 10 d后,皮质醇、丙二醛、总蛋白、谷胱甘肽过氧化物酶含量在不同处理组的稚龟中没有显著差异(P>0.05),超氧化物歧化酶含量在低温处理30 d与60 d组中有显著性差异(P<0.05),低温处理 30 d组稚龟总抗氧化能力显著高于低温处理 60 d和 90 d组(P<0.05).低温处理 60 d组稚龟单次持续游泳时间最短,为(30.61±11.74)s,显著低于低温处理 0 d组的游泳时间(P<0.05).低温处理组稚龟生长速率显著高于未处理组(P<0.05).结果表明,10℃低温保存可有效延长乌龟苗种供应时间,为兼顾稚龟存活率与品质,建议低温保存时长控制在 60 d以内.研究结果可为乌龟苗种长期保存提供具有实用价值的基础数据.
To investigate the effects of cryopreservation on the growth of Mauremys reevesii hatchlings,an artificial temperature-control method was applied.The hatchlings were preserved at 10℃for 0,30,60,and 90 days.After survival rates were recorded,the hatchlings were transferred to a rearing environment at 29℃for 10 days,then biochemical indices and locomotor performance were measured.In addition,the hatchlings subjected to low-temperature treatment were cultured at 29℃for two-month to determine growth rate.The results showed that the survival rates of the hatchlings stored at 10℃for 0,30,60,and 90 days were 100%,100%,97.78%,and 42.22%,respectively.No significant differences were observed in cortisol,malondialdehyde,total protein con-centration,or glutathione oxidase among hatchlings from different treatment groups(P>0.05).Superoxide dis-mutase activity differed significantly between the 30-day and 60-day groups(P<0.05),and the total antioxidant capacity of hatchlings in the 30-day treatment group was significantly higher than that in the 60-day and 90-day groups(P<0.05).The 60-day group exhibited the shortest single sustained swimming time(30.61±11.74)s,which was significantly lower than that of the 0-day treatment group(P<0.05).The growth rate of hatchlings in the low-temperature treatment groups was significantly higher than that of the untreated group(P<0.05).Low-temperature storage at 10℃can effectively extend the seedling supply period.To balance survival rate and quali-ty,it is recommended that the storage duration be controlled within 60 days.This study provides practical baseline data for the long-term preservation of M.reevesii hatchlings.
