目的:探讨分化簇147(cluster of differentiation 147,CD147)表达水平对乳腺癌细胞增殖、侵袭及脂代谢活性的影响.方法:收集不同分子分型的乳腺癌组织样本及对应细胞系,通过免疫组织化学、RT-PCR与Western Blot检测CD147的表达差异.分别构建CD147敲低与过表达的乳腺癌细胞模型,采用CCK-8 法、Transwell小室实验、流式细胞术以及脂质含量检测试剂盒,分析CD147对细胞增殖、侵袭、凋亡及脂代谢的影响.统计学方法采用独立样本t检验或卡方检验,以双侧P<0.05 为差异具有统计学意义.结果:乳腺癌组织中CD147 的表达水平显著高于癌旁组织(P<0.05),CD147高表达与肿瘤低分化相关(χ2=7.453,P<0.05),与淋巴结转移相关(χ2=4.554,P<0.05).生存分析显示,CD147高表达患者的总生存期较短(χ2=4.001,P<0.05).体外实验显示,CD147 过表达可促进乳腺癌细胞增殖与侵袭,并抑制细胞凋亡.此外,CD147可能通过调控脂代谢关键蛋白甾类调节元件结合蛋白 1(sterol regulatory element binding protein 1,SREBP-1)影响乳腺癌细胞的脂代谢过程.结论:CD147在乳腺癌中呈现高表达,且与不良预后相关;CD147能够促进乳腺癌细胞增殖、侵袭并抑制凋亡,其机制可能与调控SREBP-1介导的脂代谢重编程有关.

Objective:To investigate the effects of cluster of differentiation 147(CD147)expression on the proliferation,invasion,and lipid metabolism of breast cancer cells.Methods:Breast cancer tissue samples of different molecular subtypes and their corresponding cell lines were collected.Immunohistochemistry,RT-PCR,and Western blot were performed to de-tect differences in CD147 expression.CD147 knockdown and overexpression breast cancer cell models were established.Cell proliferation,invasion,apoptosis,and lipid metabolism were assessed using the CCK-8 assay,Transwell chamber assay,flow cytometry,and lipid content detection kits,respectively.Statistical analyses were conducted using the independent sam-ples t-test or chi-square test,with a two-sided P<0.05 indi-cating statistical significance.Results:CD147 expression was significantly higher in breast cancer tissues than in adjacent normal tissues(P<0.05).High CD147 expression was corre-lated with both poorer tumor differentiation(χ2=7.453,P<0.05)and lymph node metastasis(χ2=4.554,P<0.05).Survival analysis revealed that patients with high CD147 expression exhibited shorter overall survival(χ 2=4.001,P<0.05).Furthermore,CD147 overexpression promoted breast cancer cell proliferation and invasion while inhibiting apoptosis.Furthermore,CD147 may influence lipid metabolism in breast cancer cells by regulating sterol regulatory element binding protein 1(SREBP-1),a key protein involved in lipid metabolism.Conclusion:CD147 is highly expressed in breast cancer and is associated with poor prognosis.CD147 promotes proliferation and invasion while inhibiting apoptosis in breast cancer cells,and its mechanism may involve the regulation of SREBP-1-mediated lipid metabolism reprogramming.
