干露胁迫对四角蛤蜊存活率与生理生化指标的影响OA
Effects of Exposuring to Air on Survival Rate,Physiology and Biochemistry Indicators of Mactra veneriformis
试验旨在研究干露胁迫对四角蛤蜊的存活率、生理生化指标的影响,为探明四角蛤蜊干露应激反应机制、科学开展苗种培育及安全运输提供数据支撑.采用室内试验生态学方法系统研究不同干露时间(0、24、48、72、96、120 h)对四角蛤蜊成贝存活率、耗氧率和排氨率及消化酶、保护酶活性的影响,分析应激反应机制,探索适宜干露时间.结果表明:①随干露时间的延长,四角蛤蜊存活率逐渐降低,耗氧率和排氨率先升高后降低.在四角蛤蜊100%存活的干露时间内(0~72 h),耗氧率相对于干露0 h升高10%的干露时间为5.7 h,升高50%的时间为21.8 h;排氨率升高10%的干露时间为0.7 h,升高50%的时间为3.7 h.②干露胁迫后,四角蛤蜊的氧氮比(O∶N)显著低于干露0 h(P<0.05).③随着干露时间的延长,淀粉酶(amylase,AMS)活性先下降后上升;脂肪酶(lipase,LPS)活性先上升后下降;过氧化氢酶(catalase,CAT)和超氧化物歧化酶(superoxide dismutase,SOD)活性呈现出"抑制-诱导-抑制-诱导"的规律;酸性磷酸酶(acid phosphatase,ACP)活性则表现为"抑制-诱导-抑制"的规律.综上所述,干露胁迫对四角蛤蜊的存活率、生理生化方面均有一定影响,为保证四角蛤蜊的活性,在17~18℃条件下,干露时间以不超过24 h为宜,最长不要超过72 h.
In order to investigate the effects of exposure to air on survival rate,physiology and biochemistry of Mactra veneriformis,study the stress response mechanism,and provide a scientific basis for seedling rear-ing and transporting.Using an indoor experimental ecological approach,we systematically examined the impact of different air exposure durations(0,24,48,72,96 and 120 h)on the survival rate,oxygen consump-tion rate,ammonia excretion rate,digestive enzyme activities,and protective enzyme activities in Mactra veneriformis.The study aimed to analyze the stress response mechanism and determine an appropriate safe air exposure duration.The results showed that:①survival rate progressively declined with increasing expo-sure duration.In contrast,oxygen consumption and ammonia excretion rates initially increased and subse-quently decreased.Within the exposure period,maintaining 100%survival(0-72 h),the oxygen consump-tion rate increased by 10%relative to the control after 5.7 h,and by 50%after 21.8 h.Similarly,the ammonia excretion rate increased by 10%after 0.7 h and by 50%after 3.7 h.② Following air exposure stress,the oxygen-to-nitrogen ratio(O∶N)was significantly lower than that of the control group(P<0.05).③With prolonged exposure,amy-lase(AMS)activity initially decreased and then increased,while lipase(LPS)activity first increased and then decreased.Catalase(CAT)and superoxide dismutase(SOD)activities exhibited a pattern of inhibition-induction-inhibition-induction.Acid phosphatase(ACP)activity displayed an inhibition-induction-inhibition pattern.In conclusion,air exposure stress significantly impacts survival rate,physiology,and biochemistry in Mactra veneriformis.To ensure clam vitality at 17-18℃,the air expo-sure duration should ideally not exceed 24 h,and must not exceed 72 h.
