芦丁通过抑制ROS/TXNIP/NLRP3通路减轻牙周炎大鼠牙周组织损伤和炎症反应OA
Rutin alleviates periodontal tissue damage and inflammatory response in rats with periodontitis by inhibiting the ROS/TXNIP/NLRP3 pathway
目的:探讨芦丁(rutin,RU)调节活性氧(reactive oxygen species,ROS)/硫氧还蛋白相互作用蛋白(thi-oredoxin interacting protein,TXNIP)/NOD样受体热蛋白结构域相关蛋白 3(NOD-like receptor thermal protein domain associated protein 3,NLRP3)通路对牙周炎(periodontitis,PD)大鼠牙周组织损伤和炎症反应的影响.方法:将造模成功的PD大鼠随机分为PD组、RU-L组、RU-H组(灌胃 40 mg/kg、80 mg/kg的RU)、RU-H+TMAO组(灌胃 80 mg/kg的RU+110 mg/kg TMAO),每组 12 只.随机选取12只大鼠作为NC组,NC组、PD组灌胃等剂量的生理盐水,1 次/d,持续28 d.HE染色观察牙周组织病理学变化;ELISA法检测牙周组织炎症因子水平;比色法检测牙周组织氧化应激水平;DCFH-DA法检测牙周组织ROS水平;TUNEL法检测牙周组织细胞凋亡情况;Western Blot检测牙周组织相关蛋白水平.结果:与NC组相比,PD组大鼠牙周组织有大量炎症细胞浸润,伴有牙槽骨吸收、胶原纤维分解和结缔组织破坏,GI、AL值、牙周组织TNF-α、IL-1β、IL-8、IL-18、ROS、MDA水平、细胞凋亡率、TXNIP、NLRP3、ASC、Caspase 1、IL-1β、IL-18 蛋白表达显著升高(P<0.05),牙周组织SOD、CAT水平显著降低(P<0.05);与PD组相比,RU-L组、RU-H组牙周组织炎症细胞浸润、牙槽骨吸收、胶原纤维分解和结缔组织破坏程度减轻,GI、AL值、牙周组织 TNF-α、IL-1β、IL-8、IL-18、ROS、MDA水平、细胞凋亡率、TXNIP、NLRP3、ASC、Caspase 1、IL-1β、IL-18 蛋白表达显著下降(P<0.05),牙周组织SOD、CAT水平显著升高(P<0.05);与RU-H组相比,RU-H+TMAO组大鼠牙周组织破坏程度加重,GI、AL值、牙周组织TNF-α、IL-1β、IL-8、IL-18、ROS、MDA水平、细胞凋亡率、TXNIP、NLRP3、ASC、Caspase 1、IL-1β、IL-18 蛋白表达显著升高(P<0.05),牙周组织SOD、CAT水平显著降低(P<0.05).结论:RU可能通过抑制ROS/TXNIP/NLRP3 通路,减轻PD大鼠牙周组织损伤和炎症反应.
Objective:To explore the effects of rutin(RU)on periodontal tissue damage and inflammatory response in periodontitis(PD)rats by regulating the reactive oxygen species(ROS)/thioredoxin interacting protein(TXNIP)/NOD-like receptor thermal protein domain associated protein 3(NLRP3)pathway.Methods:Successful PD rats were randomly separa-ted into PD group,RU-L group,RU-H group(gavage of 40 mg/kg and 80 mg/kg RU),and RU-H+TMAO group(gavage of 80 mg/kg RU+110 mg/kg TMAO),with 12 rats in each group.Another 12 rats were stochastically selected as the NC group.The NC group and PD group were given equal doses of physiological saline by gavage,once a day for 28 days.HE staining was performed to observe pathological changes in periodontal tissue.ELISA method was used to measure the inflammatory fac-tors in periodontal tissue.Colorimetric method was performed to measure oxidative stress in periodontal tissue.DCFH-DA method was performed to detect ROS in periodontal tissue.TUNEL method was performed to measure apoptosis in periodontal tissue cells.Moreover,Western Blot was used to detect the periodontal tissue related proteins.Results:Compared with the NC group,PD group had a large amount of inflammatory cell infiltration in periodontal tissue,accompanied by alveolar bone re-sorption,collagen fiber decomposition,and connective tissue damage.The GI,AL values,periodontal tissue TNF-α,IL-1β,IL-8,IL-18,ROS,MDA levels,cell apoptosis rate,TXNIP,NLRP3,ASC,Caspase 1,IL-1β,IL-18 protein expression were clearly increased(P<0.05),and the periodontal tissue SOD and CAT were clearly decreased(P<0.05).Compared with the PD group,the RU-L and RU-H groups showed reduced infiltration of inflammatory cells,alveolar bone resorption,collagen fi-ber breakdown,and connective tissue damage in periodontal tissue.The GI,AL values,periodontal tissue TNF-α,IL-1β,IL-8,IL-18,ROS,MDA levels,cell apoptosis rate,TXNIP,NLRP3,ASC,Caspase 1,IL-1β,IL-18 protein expression were clearly decreased(P<0.05),and the periodontal tissue SOD and CAT were clearly increased(P<0.05).Compared with the RU-H group,the RU-H+TMAO group showed increased damage to periodontal tissue,the GI,AL values,periodontal tissue TNF-α,IL-1β,IL-8,IL-18,ROS,MDA levels,cell apoptosis rate,TXNIP,NLRP3,ASC,Caspase 1,IL-1β,IL-18 protein expression were clearly increased(P<0.05),and the periodontal tissue SOD and CAT were clearly decreased(P<0.05).Conclusion:RU may alleviate periodontal tissue damage and inflammatory response in PD rats by inhibiting ROS/TXNIP/NLRP3 pathway.
