[目的]对感染黄龙病的赣南脐橙与健康的赣南脐橙内生细菌和根际土壤细菌微生物的多样性和群落组成进行了研究,探究染病与健康条件下赣南脐橙内生细菌和根际土壤细菌微生物物种组成和群落结构的变化规律.[方法]以赣南脐橙为研究对象,采集患黄龙病植株和未发病植株的叶片、果实、根、根际土壤样品开展16S rDNA扩增子测序,利用高通量测序技术获取特定试验样本中细菌的物种组成、群落丰度、群落差异等多方面信息.[结果]在健康和感病脐橙中,根际土壤细菌的OTUs最多,其次是根部、叶片,果实最少;健康脐橙果实、叶片、根部内生细菌和根际土壤细菌的Shannon指数值均大于感病样品,表明健康脐橙果实、叶片、根部内生细菌和根际土壤细菌的物种多样性均高于感病脐橙;健康和感病的脐橙内生细菌在门水平上主要有蓝菌门(Cyanobacteria),其中根部蓝菌门的相对丰度最低,叶片蓝菌门的相对丰度最高;在健康和感病的脐橙根际土壤细菌中,假单胞菌门(Pseudomonadota)、放线菌门(Actinomycetota)、酸杆菌门(Acidobacteriota)、绿弯菌门(Chloroflexota)为主要优势细菌门.[结论]赣南脐橙感病和健康的叶片、果实、根内生细菌以及根际土壤细菌群落多样性丰富,存在相关的有益细菌,为今后探索赣南脐橙内生细菌及其根际土壤细菌群落多样性的利用途径,以及柑橘黄龙病的绿色防控研究,提供了新的参考依据.

[Objective]To investigate the diversity and community composition of endophytic bacteria and rhizosphere soil bacterial microorganisms in Gannan Navel Oranges infected with Huanglong disease and healthy Gannan Navel Oranges,and to explore the changes in species composition and community structure of endophytic bacteria and rhizosphere soil bacterial microorganisms in Gannan Navel Oranges under disease and non disease conditions.[Method]Taking Gannan Navel Orange as the research object,leaf,fruit,root,and rhizosphere soil samples of Huanglongbing and non diseased plants were collected for 16S rDNA amplicon sequencing.High throughput sequencing technology was used to obtain various information such as bacterial species composition,community abundance,and community differences comparison in specific experimental samples.[Result]Among healthy and diseased navel oranges,the OTUs of rhizosphere soil bacteria were the highest,followed by roots and leaves,and the lowest in fruits;The Shannon index values of endophytic bacteria and rhizosphere soil bacteria in healthy navel orange fruits,leaves,and roots were higher than those in susceptible samples,indicating that the species diversity of endophytic bacteria and rhizosphere soil bacteria in healthy navel orange fruits,leaves,and roots was higher than that in susceptible navel oranges;The endophytic bacteria in healthy and diseased navel oranges mainly include Cyanobacteria at the phylum level,with the lowest relative abundance of Cyanobacteria in roots and the highest relative abundance of Cyanobacteria in leaves;Among the bacteria in the rhizosphere soil of healthy and diseased navel oranges,Pseudomonas,Actinomycota,Acidobacteriota,and Chloroflexita are the main dominant bacterial phyla.[Conclusion]The diversity of endophytic bacteria and rhizosphere soil bacterial communities in susceptible and healthy leaves,fruits,roots,and rhizosphere soil of Gannan Navel Oranges is abundant,and there are related beneficial bacteria.This provides new reference for exploring the utilization pathways of endophytic bacteria and rhizosphere soil bacterial communities in Gannan Navel Oranges in the future,as well as green prevention and control research on citrus huanglongbing.
