桔褐天牛为害与寄主营养和次生代谢物质的相关性分析OA
Correlation analysis between the damage of Nadezhdiella cantori and host plant nutrition and secondary metabolites
[目的]调查了解桔褐天牛(Nadezhdiella cantori)在江西樟树药材基地对樟头红的为害情况,分析其为害与寄主营养和次生代谢物质之间的相互关系,为探索桔褐天牛生态调控技术提供参考.[方法]通过样线调查法明确桔褐天牛对樟头红的为害状况,采用蒽酮比色法、3,5-二硝基水杨酸法(DNS法)、考马斯亮蓝G-250法分别测定樟头红树干胸径和基部可溶性总糖、还原糖和可溶性蛋白含量,采用分光光度法测定多酚、单宁、黄酮和总三萜含量,分析桔褐天牛为害与樟头红树干营养和次生代谢物质含量之间的关系.[结果]樟树市桔褐天牛对樟头红的为害率为 36.15%±5.89%,桔褐天牛蛀孔 70%以上集中于樟头红树干胸径及以上部位.樟头红树干营养物质含量总体表现为可溶性总糖>还原糖>可溶性蛋白质.可溶性总糖及可溶性蛋白质含量在树干胸径处和基部之间无显著差异,胸径处的还原糖含量则显著高于基部;桔褐天牛为害与樟头红树干还原糖含量呈显著正相关,与其他部位营养物质无显著相关性.多酚在樟头红树干次生代谢物质中含量较高,桔褐天牛为害与樟头红胸径处的多酚、总三萜及不同部位单宁含量均呈负相关.单宁和总三萜含量在桔褐天牛为害的虫伤株中普遍下降,多酚含量在虫伤株中的树干基部显著增加.[结论]樟头红树干还原糖含量对桔褐天牛为害选择具有重要影响,桔褐天牛的蛀孔危害可导致寄主植物树干的总三萜和单宁含量降低,多酚在桔褐天牛寄主植物防御体系中发挥主导作用.
[Objective]The damage of Nadezhdiella cantori to Zhangtouhong in Jiangxi's Zhangshu medicinal materials base was investigated,and the relationship between its damage and nutrients and secondary metabolites of host plants was analyzed.The study provides references for exploring the ecological regulation technology of N.cantori.[Method]The damage status of N.cantori to Zhangtouhong was investigated by sampling line survey.The total soluble sugar,reducing sugar and soluble protein content at DBH(diameter at breast height)and base in the trunk of Zhangtouhong were measured by using anthrone colorimetry,the 3.5-dinitrosalicylic acid(DNS)method and Coomassie Brilliant Blue G-250 assay,respectively.The content of polyphenols,tannins,flavonoids and total triterpenes was determined by spectrophotometry,and the relationship between the damage of N.cantori and the content of nutrition and secondary metabolites of Zhangtouhong was analyzed.[Result]The damage rate of Zhangtouhong by N.cantori was 36.15±5.89%in Zhangshu City,and over 70%of the borer holes of N.cantori were concentrated at or above the DBH of the Zhangtouhong trunk.The content of nutrients in the trunk of Zhangtouhong generally followed the order:total soluble sugar>reducing sugar>soluble protein.There was no significant difference in the content of soluble total sugar and soluble protein between the DBH and the base,while the content of reducing sugar in the DBH was significantly higher than that in the base.The damage of N.cantori was significantly positively correlated with the content of reducing sugar in Zhangtouhong trunk,but not with nutrients in other parts.Polyphenols were secondary metabolites with high content in the trunk of Zhangtouhong,and the damage of N.cantori was negatively correlated with the content of polyphenols,total triterpenes at DBH and tannin in different parts.The content of tannin and total triterpenoids decreased in the plants damaged by N.cantori,while the content of polyphenols increased significantly in the base of the trunk.[Conclusion]The content of reducing sugar of Zhangtouhong has an important influence on the pest selection of N.cantori.The damage from borer holes could reduce the content of total triterpenoids and tannins in the trunk of the host plants of N.cantori,and polyphenols play a leading role in the defense system of host plants of N.cantori.
