白鲜碱对6种植物病原真菌的抗菌活性评价OA

A study on antifungal activities of dictamnine against six plant pathogenic fungi

[目的]多种植物次生代谢产物,如香豆素、生物碱和黄酮类化合物等,具有良好的抗菌活性,是开发植物源杀菌剂的优质潜在资源.实验室长期致力于黄皮等特色植物中活性成分的挖掘和利用工作,期望获得更多的绿色源抗菌化合物.[方法]以黄皮植物茎叶为材料,采用多种柱层析和半制备型液相等技术进行分离纯化,通过 1 H NMR、13 C NMR等波谱技术鉴定化合物结构,得到一个呋喃喹啉生物碱成分白鲜碱.采用菌丝生长速率法研究了白鲜碱对 6种常见植物病原真菌的抗菌活性.[结果]白鲜碱对 6 种植物病原菌的菌丝生长均表现出较好的抑制效果,且呈现随着化合物浓度升高,抗菌活性增强的趋势.白鲜碱对茄镰孢菌(Fusarium solani)、尖孢镰孢菌(F.oxysporum)、层出镰孢菌(F.proliferatum)、禾谷镰孢菌(F.graminearum)、葡萄座腔菌(Botryosphaeria dothidea)和灰葡萄孢菌(Botrytis cinerea)的EC50 值分别为92.61、39.37、33.09、9.9、15.48 和 10.81 μg/mL.其中白鲜碱对禾谷镰孢菌、葡萄座腔菌和灰葡萄孢菌的抑制效果较显著.对白鲜碱处理的病菌菌丝形态进行观察,发现白鲜碱处理后病原菌菌丝出现扭曲、肿胀和空腔等特征变化.[结论]白鲜碱对 6 种植物病原真菌的生长和形态均有一定影响,具有广谱抗性.

[Objective]Botanical fungicides have become one of the ideal choices for the research and development of green fungicides due to their unique environmental-friendly characteristics,biodegradability,and efficient selectivity for specific targets.Plant secondary metabolites,such as coumarins,anthocyanins,phenols,alkaloids,terpenoids and flavonoids,etc.,exhibit excellent antifungal activities and become the main sources for the development of botanical fungicides.[Method]In this study,the stems and leaves of Clausena lansium were used as experimental materials.Separation and purification were carried out by various column chromatography and semi-preparative liquid chromatography techniques.The structure of the compound was identified by spectroscopic techniques such as 1 H NMR and 13 C NMR,and a furanocoline alkaloid component,dictamnine,was obtained.The antifungal activity of dictamnine against six kinds of phytopathogenic fungi was tested by the mycelial growth rate method.[Result]The results showed that dictamnine inhibited the mycelial growth and morphology of six plant pathogenic fungi.The EC50 values of Fusarium solani,F.oxysporum,F.proliferatum,F.graminearum,Botryosphaeria dothidea and Botrytis cinerea were 92.61,39.37,33.09,9.9,15.48 and 10.81 μg/mL,respectively.Among them,dictamnine had a significant inhibitory effect on F.gramineum,B.dothidea and B.cinerea.Observation of the mycelial morphology of the pathogen treated with dictamnine revealed that the mycelia of the pathogen showed characteristic changes such as twisting,swelling and cavities after treatment with dictamnine.[Conclusion]It indicates that dictamnine has certain effects on the growth and morphology of six plant pathogenic fungi and possesses broad-spectrum resistance.
