金合欢素对酒精诱导小鼠肝损伤的保护作用OA
Protective effect of acacetin against alcohol-induced liver injury in mice
探讨金合欢素(Acacetin,ACA)对酒精诱导的小鼠肝脏组织损伤的影响.酒精灌胃诱导肝损伤后处死小鼠,称量体质量和肝脏质量,通过HE染色观察肝脏形态学变化,试剂盒检测小鼠血清中谷丙转氨酶(Alanine aminotransferase,ALT)活性,并检测炎症因子相关基因IL-6,IL-10,TNFα 和抗氧化相关基因 SOD,Nrf2,HO-1,NQO1 的 mRNA 表达水平.结果显示,与对照组(CK)相比,模型组(Model)小鼠血清ALT活性显著升高,炎症因子相关基因mRNA表达水平上调,抗氧化酶相关基因 mRNA 表达水平下降.与 Model 组相比,金合欢素低剂量干预组(Model+LACA)和高剂量干预组(Model+HACA),小鼠血清ALT活性显著降低,炎症因子相关基因mRNA表达水平降低,抗氧化酶相关基因mRNA表达水平均升高.结果表明,ACA能够减轻酒精诱导的小鼠肝脏损伤,研究结果可为ACA在酒精性肝病防治中的应用提供实验依据.
Aims to investigate the effects of acacia saponin on alcohol-induced liver tissue damage in mice.After inducing liver injury through alcohol gavage,mice were euthanized,body mass and liver mass were measured,liver morphological changes were observed via HE staining,alanine aminotransferase activity in serum was detected using a kit,and mRNA expression levels of inflammatory factor-related genes IL-6,IL-10,TNFα and antioxidant-related genes SOD,Nrf2,HO-1,NQO1 were assessed.Results showed that compared with the control group(CK)mice in the model group(Model)exhibited significantly elevated serum ALT activity,upregulation of mRNA expression levels for pro-inflammatory genes,and downregulation of mRNA expression levels for antioxidant-related genes.Compared with the Model group,both the low-dose acacia-lactone intervention group(Model+LACA)and the high-dose intervention group(Model+HACA)exhibited significantly reduced ALT activity,decreased mRNA expression levels of pro-inflammatory genes,and increased mRNA expression levels of antioxidant enzyme genes.The results indicate that acacia reticulata extract alleviates alcohol-induced liver injury in mice.These findings provide experimental evidence supporting the application of acacia reticulata extract in the prevention and treatment of alcoholic liver disease.
