旨在探究橙皮素(HPT)对脂多糖(LPS)诱导的人肠上皮细胞(NCM460)氧化损伤及凋亡的影响.采用 MTT 法检测不同浓度 HPT 对 NCM460 细胞活力的影响,确定 50,200,400 μmol/L HPT用于后续实验.实验分为空白对照组、LPS单独处理组和不同浓度HPT与LPS共处理组.利用生化试剂盒检测各组细胞中丙二醛(MDA)和活性氧(ROS)水平、谷胱甘肽过氧化物酶(GSH-Px)活性、超氧化物歧化酶(SOD)活性、总抗氧化能力(T-AOC)以及Caspase3/9活性的影响.利用荧光定量PCR技术检测抗氧化和细胞凋亡相关基因mRNA的相对表达量.结果显示,与 LPS 处理组相比,不同浓度 HPT 预处理后,细胞中 MDA 和 ROS 水平显著降低,GSH-Px活性、SOD活性和T-AOC显著升高,Caspase3/9 活性显著降低;抗氧化相关基因Nrf2,HO-1,NQO1,SOD1和抗凋亡因子Bcl-2 mRNA的相对表达量显著升高;促凋亡因子Caspase3/9和 Bax mRNA 的相对表达量显著降低.研究表明,适宜浓度的 HPT 能够有效抑制 LPS 诱导的NCM460 细胞的氧化损伤和细胞凋亡.

Aimed to investigate the effects of Hesperetin(HPT)on oxidative damage and apoptosis in lipopolysaccharide(LPS)-induced human intestinal epithelial cells(NCM460).Cell viability of NCM460 cells was assessed using the MTT assay to determine the optimal concentration of HPT,and finally 50,200,400 μmol/L HPT were selected for subsequent experiments.The experimental groups included the control group,LPS group and different concentrations of HPT+LPS group.Biochemical assays were conducted to measure malondialdehyde(MDA)and reactive oxygen species(ROS)levels,glutathione peroxidase(GSH-Px)activity,superoxide dismutase(SOD)activity,total antioxidant capacity(T-AOC),as well as Caspase3/9 activity in the cells.The relative expression levels of antioxidant and apoptosis-related genes mRNA were determined using fluorescence quantitative PCR.The results showed that pretreatment with different concentrations of HPT significantly reduced MDA and ROS levels,increased GSH-Px,SOD activity,T-AOC and decreased Caspase3/9 activity compared to the LPS group.The relative expression levels of antioxidant-related genes Nrf2,HO-1,NQO1,SOD1 and anti-apoptotic factor Bcl-2 mRNA were significantly upregulated,while pro-apoptotic factors Caspase3/9 and Bax mRNA were significantly downregulated.These findings indicate that appropriate concentrations of HPT can effectively inhibit oxidative damage and apoptosis in LPS-induced NCM460 cells.
