[目的]探究影响粉葛和野葛间淀粉积累特异性的关键因素,为粉葛转录调控和分子育种提供理论基础.[方法]收集了6份药食同源葛根资源,分析1年生葛根的形态与淀粉粒细胞差异,开展差异转录组分析,识别差异表达基因(DEGs);进一步通过相关性分析筛选与淀粉积累显著相关基因,挖掘影响淀粉积累特异性的潜在关键基因.[结果]淀粉含量最高的为粉葛(G1),其含量为野葛的2倍,且粉葛次生结构组织细胞发育与扩张更快;差异转录组分析共鉴定出4718个DEGs,主要涉及生物调节、细胞膜结构、细胞外区域、胞间连丝等结构以及转录因子和信号转导等功能类别;同时,发现137个与淀粉积累显著正相关基因和231个负相关基因,其中9个参与淀粉和糖代谢的基因及12个转录因子被识别为影响淀粉积累特异性的潜在关键基因集.[结论]构建了葛根比较转录组数据库,挖掘并分析了影响其淀粉积累特异性的关键差异基因集,为粉葛的转录调控机制解析及分子育种实践提供了理论依据.

[Objective]This study aimed to investigate the key factors underlying the differential starch accumulation between Pueraria montana var.thomsonii(P.thomsonii)and Pueraria montana var.lobata(P.lobata),to provide a theoretical basis for the molecular breeding of high-starch kudzu varieties.[Method]Six medicinal and edible kudzu resources were collected.The morphological and starch granule characteristics of one-year-old roots were analyzed.Differential transcriptome analysis was performed to identify differentially expressed genes(DEGs),and correlation analysis was conducted to pinpoint genes associated with starch accumulation.[Result]The P.thomsonii accession G1 exhibited the highest starch content,which was twice that of P.lobata,and demonstrated more rapid development and expansion of secondary structural tissue cells.Transcriptome analysis revealed 4718 DEGs,predominantly enriched in functional categories including biological regulation,cellular membrane structures,extracellular regions,plasmodesmata,transcription factors,and signal transduction.Furthermore,137 and 231 genes were significantly positively and negatively correlated with starch accumulation,respectively.Among these,nine genes involved in starch and sugar metabolism and twelve transcription factors were identified as potential key regulators of starch accumulation specificity.[Conclusion]This study establishes a comparative transcriptome database and identifies critical gene set influencing starch accumulation,thereby providing a solid theoretical foundation for the transcriptional regulation and molecular breeding of P.thomsonii.
