为探究C型凝集素(CTL)基因家族对舞毒蛾(Lymantria dispar)抗球孢白僵菌(Beauveria bassiana)能力的影响,对舞毒蛾CTL家族进行了生物信息学分析鉴定,发现大部分CTL含两个CTLD保守结构域,其中,舞毒蛾CTL-10 蛋白与已鉴定的鳞翅目(Lepidoptera)CTL亲缘关系较近;舞毒蛾被球孢白僵菌侵染后,体内CTL基因表达在 48 h内显著升高;并且在抑制CTL-10 后 96 h,球孢白僵菌对舞毒蛾的致死率提高 26%.进一步检测舞毒蛾细胞免疫能力,敲低CTL-10 显著降低了舞毒蛾血细胞总数与包裹能力;除此之外,球孢白僵菌侵染可以诱导舞毒蛾Toll通路基因表达,敲低CTL-10 显著降低球孢白僵菌对上述基因的诱导.结果证明,CTL-10 是舞毒蛾抗球孢白僵菌入侵的重要基因,通过CTL-10 抗真菌能力分析有利于为舞毒蛾绿色生物防治提供新靶标.

To investigate the effects of the C-type lectin(CTL)gene family on the resistance of Lymantria dispar to Beauveria bassiana,a comprehensive bioinformatic analysis and identification of the L.dispar CTL family were performed.The results showed that most CTL contained two CTLD conserved domains.Among them,the L.dispar CTL-10 protein exhibited a close phylogenetic relationship with the previously identified CTLs from Lepidoptera species.After infection with B.bassi-ana,the expression levels of CTL genes in L.dispar were significantly up-regulated within 48 hours.Furthermore,after in-hibiting CTL-10 for 96 hours,it resulted in a 26%increase in the mortality rate of L.dispar caused by B.bassiana.The cellular immune capacity of L.dispar was further detected.The knockdown of CTL-10 significantly reduced the total hemo-cyte count and encapsulation ability of L.dispar.In addition,B.bassiana infection could induce the expression of genes in the Toll signaling pathway of L.dispar,while the knockdown of CTL-10 significantly attenuated this inductive effect of B.bassiana on the aforementioned genes.Collectively,these results demonstrated that CTL-10 is a crucial gene involved in the resistance of L.dispar to B.bassiana invasion.The analysis of the antifungal activity of CTL-10 will facilitate the iden-tification of novel targets for the green biological control of L.dispar.
