依据表型指标和分子标记的红松种子园遗传多样性分析OA
Genetic Diversity Analysis of Pinus koraiensis Seed Orchards Based on Phenotypic Traits and Molecular Mark-ers
为明确红松种子园的遗传背景,以辽宁省森林经营研究所实验林场红松一代无性系种子园 24 个无性系为材料,通过分析13 个表型指标在各红松无性系间差异与多样性,并利用SSR标记技术从分子水平分析红松种子园遗传多样性,进而构建各红松无性系的分子身份证.结果表明:红松种子园在表型和分子水平上均存在较为丰富的遗传多样性.13 个表型指标变异系数介于 7.48%~91.77%,均值为 23.67%,其中,生长性状和种实性状的变异系数均值分别为 18.90%、31.30%;Shannon-Weaver多样性指数介于 0.928~2.084,均值为 1.908,生长性状和种实性状的多样性指数均值分别为 1.890、1.938,说明红松种子园不同无性系生长性状数值的高低差异较大,在种实性状上数值分级更为丰富.在分子标记结果中,10 对SSR引物在 24 个红松无性系中共检测出 43 个等位基因,有效等位基因数、Shannon信息指数、期望杂合度、观测杂合度均值分别为 2.207、0.906、0.476、0.421;表型和SSR聚类均将红松种子园内无性系分为 3 大类群,但类群内系号差异较大,并通过 3 对高效引物构建了 24 个红松无性系的分子身份证,分辨率达 100%.
To clarify the genetic background of Pinus koraiensis seed orchard,24 clones from the first-generation clonal seed or-chard of P.koraiensis at the Experimental Forest Farm of Liaoning Provincial Institute of Forest Management were selected as research materials.We analyzed the differences and diversity of 13 phenotypic traits among P.koraiensis clones,and evaluated the genetic diversity of the seed orchard at the molecular level using SSR marker technology,thereby constructing molecular identity cards for each P.koraiensis clone.The results showed that the P.koraiensis seed orchard exhibited abun-dant genetic diversity at both phenotypic and molecular levels.The coefficient of variation(CV)of the 13 phenotypic traits ranged from 7.48%to 91.77%,with an average of 23.67%.Among them,the mean CV values of growth traits and seed traits were 18.90%and 31.30%,respectively.The Shannon-Weaver diversity index varied from 0.928 to 2.084,with an average of 1.908,and the mean diversity index values of growth traits and seed traits were 1.890 and 1.938,respectively.These findings indicated that the growth traits of different clones in the seed orchard showed large numerical differences,while the seed traits had a more abundant numerical classification.In terms of molecular marker results,a total of 43 al-leles were detected in the 24 P.koraiensis clones using 10 pairs of SSR primers.The mean values of the effective number of alleles,Shannon's information index,expected heterozygosity,and observed heterozygosity were 2.207,0.906,0.476,and 0.421,respectively.Cluster analysis based on both phenotypic traits and SSR markers divided the clones in the seed orchard into three major groups,but there were significant differences in clone numbers within each group.Furthermore,molecular identity cards for the 24 P.koraiensis clones were constructed using 3 pairs of high-efficiency primers,with a resolution rate of 100%.
