猫泛白细胞减少症(Feline panleukopenia)是严重危害猫科动物健康的一种高度接触性传染病,其病原为猫泛白细胞减少症病毒(FPV).采集临床中疑似FPV感染猫的病料,处理后接种猫肾细胞(cran-dell reese feline kidney,CRFK),盲传至第4代出现明显细胞病变,PCR及荧光定量PCR鉴定为FPV阳性,电镜观察可见无囊膜、直径20~25 nm、球形的病毒粒子,与FPV典型形态特征一致,将其命名为FPV-pA.基于FPV VP2基因同源性分析及遗传进化分析发现,FPV-pA株与FPV Cu-4株的同源性为99.5%,与比对毒株的同源性在98.0%~100%,与FPV-QD22-8株、FPLV-CC-19-02株及cat-1株亲缘关系较近.基于全基因组同源性分析及遗传进化分析发现,FPV-pA株与FPV Cu-4株的同源性为99.3%,与比对毒株的同源性在97.5%~99.8%,与FPV-QD22-8株亲缘关系较近.动物攻毒试验结果表明,经口服104.5 TCID50病毒可使3月龄健康猫出现典型FPV感染症状.本研究成功分离并鉴定了1株FPV,该分离株对猫具有致病性,为后续研发猫泛白细胞减少症疫苗和临床治疗药物奠定了基础.

Feline panleukopenia is a highly contagious infectious disease that seriously endangers the health of felines,and its pathogen is feline panleukopenia virus.The purpose of this study was to isolate and iden-tify a strain of FPV and explore its pathogenicity.Collected clinical materials suspected of FPV infection were inoculated into Crandell Reese Feline Kidney(CRFK)cells.Obvious cytopathic effects occurred when CRFK cells were blindly passaged to the fourth generation.It was identified as FPV-positive by PCR and qPCR.Electron microscopy revealed a no envelope,20-25 nm diameter,spherical virus particle,which was consistent with the typical morphological characteristics of FPV.The isolated viral strain was named FPV-pA.Based on homology analysis and phylogenetic analysis of the FPV VP2 gene,it was found that the ho-mology between FPV-pA and FPV Cu-4 strain was 99.5%,and the homology with the matched strain was 98.0%-100%.The FPV-pA has a relatively close genetic relationship with FPV-QD22-8,FPLV-CC-19-02 and cat-1 strains.Based on genome-wide homology analysis and phylogenetic analysis,it was found that the homology between FPV-pA and FPV Cu-4 strain was 99.3%,and the homology with the matched strain was 97.5%-99.8%.The FPV-pA has a relatively close genetic relationship with FPV-QD22-8 strain.The TCID50 of the FPV-pA strain was 10-4.5CFU/mL.The results of animal challenge test showed that oral ad-ministration of 104.5 TCID50 the virus could cause typical symptoms of FPV infection in healthy three-month-old cats.In this study,a pathogenic clinical strain of FPV was successfully isolated and identified.It laid the foundation for the subsequent development of vaccines and clinical therapeutic drugs against feline panleukopenia.
