动物模型是将治疗方法从实验室转化到临床应用的重要工具.随着基因编辑技术的不断发展,基于成簇规则间隔短回文重复序列/CRISPR相关蛋白9(clustered regularly interspaced short palindromic repeats/CRISPR associated protein 9,CRISPR/Cas9)构建的动物模型为研究发病机制、开发新的治疗方法以及评估药物疗效提供了新的可能.论文简述了CRISPR/Cas9相关技术及原理,总结了基因编辑动物模型在人类和动物转化医学研究中的应用,探讨了CRISPR/Cas9应用过程中面临的挑战和解决策略,以期促进基因编辑技术在动物模型构建中的应用.
Animal models are important tools for translating therapies from the laboratory to clinical appli-cations.With the continuous development of gene editing technology,construction of gene editing animal models based on clustered regularly interspaced short palindromic repeats/CRISPR associated protein 9(CRISPR/Cas9)offers new possibilities for studying pathogenesis,developing new therapeutic approaches,and evaluating drug efficacy.This paper briefly describes technology and principles of CRISPR/Cas9,sum-marises the application of gene editing animal models in human and animal translational medicine research,and discusses the challenges and solution strategies during the application of CRISPR/Cas9,in order to promote the application of gene editing technology in the construction of animal models.
