荷叶多糖NFP-0.8-b的纯化、磷酸化修饰及其生物活性OA
Purification,Phosphorylation Modification and Biological Activity of a Novel Polysaccharide NFP-0.8-b from Nelumbinis Folium
以荷叶为原料,以去离子水为溶剂提取荷叶多糖(Polysaccharide from Nelumbinis Folium NFP),经过DEAE-Sepharose Fast Flow阴离子交换柱和Sephadex G-100凝胶色谱柱纯化得到荷叶多糖NFP-0.8-b.在此基础上,优化确定NFP-0.8-b的最佳磷酸化修饰工艺,并探讨磷酸化NFP-0.8-b(Phosphorylated NFP-0.8-b,Ph-NFP-0.8-b)的抗氧化活性及其在曲奇生产过程中对晚期糖基化末端产物的抑制作用.结果表明:NFP-0.8-b磷酸化最佳工艺为:磷酸化试剂比例(三聚磷酸钠︰三偏磷酸钠)为5︰2,反应温度75℃,pH 9.0,反应时间3.5 h,在此条件下磷酸根含量达8.39%.与NFP-0.8-b相比,Ph-NFP-0.8-b对DPPH自由基和羟基自由基的清除作用进一步增强.多糖添加量大于5%时,Ph-NFP-0.8-b对曲奇生产过程中晚期糖基化末端产物的抑制作用明显强于NFP-0.8-b(P<0.05),揭示Ph-NFP-0.8-b可以作为抗氧化剂及晚期糖基化末端产物抑制剂应用于曲奇的工业化生产.
Polysaccharides from Nelumbinis Folium(NFP)were extracted with deionized water as the solvent.A novel polysaccharide NFP-0.8-b was purified by DEAE-Sepharose Fast Flow anion exchange column and Sephadex G-100 gel chromatography column.On this basis,the optimal phosphorylation conditions for NFP-0.8-b were determined.The antioxidant activity of phosphorylated NFP-0.8-b(Ph-NFP-0.8-b)and its inhibitory effect on advanced glycation end products during cookie production were investigated.The results show that the optimal phosphorylation process of NFP-0.8-b is as follows:phosphorylation reagent ratio(sodium tripolyphosphate︰sodium trimetaphosphate)5︰2,reaction temperature 75℃,pH 9.0,and reaction time 3.5 h.Under these conditions,the phosphate content reaches 8.39%.Compared with NFP-0.8-b,the scavenging effect of Ph-NFP-0.8-b on DPPH free radicals and hydroxyl free radicals are further enhanced.When the polysaccharide content is greater than 5%,the inhibitory effect of Ph-NFP-0.8-b on advanced glycation end products during cookie production is significantly stronger than that of NFP-0.8-b(P<0.05),suggesting that Ph-NFP-0.8-b could be used as an antioxidant and inhibitor of advanced glycation end products in the industrial production of cookies.
