为探究海带多糖的最佳提取工艺,并对其结构特性及抗氧化活性进行分析,采用微波辅助低共熔溶剂(deep eutectic solvent,DES)提取海带多糖,通过单因素试验和响应面试验获得最佳提取工艺,利用傅里叶变换红外光谱、X-射线衍射和场发射扫描电子显微镜等对其进行结构表征,同时对其抗氧化活性进行分析.结果表明,海带多糖的最优提取工艺为含水质量分数20%的DES(甜菜碱与尿素摩尔比1∶2)、固液比1∶40(g/mL)、微波功率426 W、提取温度56℃、提取时间60 min,在此条件下获得多糖质量分数为5.96%,比热水浸提法提高52.04%;傅里叶变换红外光谱分析显示,微波辅助DES提取的海带多糖结构具有多样性;基于X-射线衍射分析,海带多糖为非结晶物质;场发射扫描电镜显示,热水浸提、微波辅助DES提取对海带多糖结构有不同影响.微波辅助甜菜碱-尿素提取的海带多糖具有较好的抗氧化活性,对ABTS+·自由基、DPPH自由基清除率分别为83.40%,62.34%.研究为开发和优化海带多糖的高值化利用提供理论依据.

To explore the optimal extraction process for laminaria japonica polysaccharides(LJP)and to analyze their structural characteristics and antioxidant activity,a microwave-assisted deep eutectic solvent(DES)extraction method was employed.The optimal extraction process was obtained through single-factor experiments and response surface methodology.The extracted polysaccharides were structurally characterized using Fourier transform infrared spectroscopy(FT-IR),X-ray diffraction(XRD),and field emission scanning electron microscopy(FE-SEM),and their antioxidant activity was analyzed.The results showed that the optimal extraction conditions for LJP were:DES with 20%water content(betaine to urea molar ratio of 1:2),solid-to-liquid ratio of 1:40(g/mL),microwave power of 426 W,extraction temperature of 56℃,and extraction time of 60 min.Under these conditions,the polysaccharide yield reached 5.96%,which was 52.04%higher than that obtained by hot water extraction.FT-IR analysis indicated that the structure of LJP extracted by microwave-assisted DES was diverse.XRD analysis revealed that LJP was an amorphous substance.FE-SEM showed that hot water extraction and microwave-assisted DES extraction had different effects on the structure of LJP.The LJP extracted by microwave-assisted betaine-urea exhibited good antioxidant activity,with ABTS+• and DPPH radical scavenging rates of 83.40%and 62.34%,respectively.This study provides a theoretical basis for developing and optimizing the high-value utilization of laminaria japonica polysaccharides.
