黑果枸杞LrWRKY44的克隆、生物信息学及差异表达分析OA
Cloning,Bioinformatics and Expression Analysis of LrWRKY44 in Lycium ruthenicum Murr.
以黑果枸杞的果实、根、茎、叶、花为试材,通过生物信息学工具分析了黑果枸杞LrWRKY44的结构,并通过RT-qPCR分析黑果枸杞在不同组织及不同发育时期的表达模式,以期初步探究LrWRKY44在黑果枸杞生长发育中的生物学功能.结果表明:LrWRKY44在黑果枸杞的根、茎、叶、花及果实中均有表达,其中在果实中的表达量最高.随着果实的发育,LrWRKY44的表达量逐渐增加,在成熟期达到峰值.这一表达模式与花青素含量的积累趋势相吻合.LrWRKY44可能与黑果枸杞花青素的生物合成与积累密切相关,对黑果枸杞果实色泽形成具有重要调控作用.
Taking Lycium ruthenicum fruit,root,stem,leaf,and flower as the experimental materials,the structure of LrWRKY44 was analyzed using bioinformatics tools,and its expression patterns in different tissues and at different developmental stages were determined by RT-qPCR,in order to preliminarily explore its biological function in the growth and development of L.ruthenicum.The results showed that LrWRKY44 was expressed in all parts of L.ruthenicum,including roots,stems,leaves,flowers,and fruits,with the highest expression level found in fruits.As the fruit matures,the expression of LrWRKY44 gradually increases,peaking at maturity,which aligns with the accumulation trend of anthocyanin content.The results indicated that LrWRKY44 might be closely related to the biosynthesis and accumulation of anthocyanins in L.ruthenium and played an important regulatory role in the formation of fruit color.
