菜豆复合诱变突变体库构建及基因组分析OA
Construction of a Composite Mutagenized Snap Bean Mutant Library and Genomic Analysis
以矮生食荚菜豆A18种子为试材,采用氮离子(4×1016 ions·cm-2)和60Co-γ射线(80 Gy)复合诱变的方法,研究了复合诱变对菜豆表型变异的影响及其突变体的基因组特征,以期为菜豆遗传育种与功能基因组学研究创制基础材料,也为同类作物复合诱变研究提供参考依据.结果表明:M2代共筛选获得叶、花/育性、茎/株型、籽粒和生育期突变等五大类突变体374株,其突变频率达6.34%.选取9份稳定遗传的突变体繁殖至M3代,进行全基因组重测序分析,发现突变体的核苷酸多样性(Pi:0.184~0.195)与遗传分化系数(Fst:0.111~0.172)存在明显差异,且突变体与野生型的遗传相似性系数为 0.83~0.87.
Taking seeds of dwarf edible-podded snap bean(Phaseolus vulgaris)A18 as experimental material,a combined mutagenesis method involving nitrogen ion beam(4 × 1016 ions·cm-2)and 60Co-γ ray(80 Gy)irradiation was applied to investigate the effects of composite mutagenesis on phenotypic variation in snap bean and the genomic characteristics of the mutants,in order to create foundational materials for genetic breeding and functional genomics research in snap bean and provide reference for composite mutagenesis research in related crops.The results showed that a total of 374 mutant plants belonging to five phenotypic categories-leaf,flower/fertility,stem/plant architecture,seed,and growth period were screened in the M2 generation,with a mutation frequency of 6.34%.Nine stably inherited mutant lines were selected,advanced to the M3 generation,and subjected to whole-genome resequencing analysis.Significant differences were observed among mutants in nucleotide diversity(Pi:0.184-0.195)and genetic differentiation coefficient(Fst:0.111-0.172).The genetic similarity coefficients between mutants and the wild type ranged from 0.83 to 0.87.
