The Effect of Tangeretin on Interleukin-1β-Induced Chondrocyte Damage by Regulating the AMPK/SIRT1/NF-κB Signaling Pathway

目的:探究橘红素对白细胞介素-1β(IL-1β)诱导的软骨细胞损伤的影响,分析其机制与AMP激活蛋白激酶(AMPK)/沉默信息调节因子2相关酶1(SIRT1)/核因子κB(NF-κB)通路的关系.方法:培养软骨细胞,并分为对照组、白细胞介素-1β组、低/中/高剂量橘红素组、高剂量橘红素+AMPK抑制剂Compound C组(高剂量橘红素+Compound C组).用克隆形成实验、流式细胞术、DCFH-DA荧光探针、Western Blot法分别检测细胞增殖、凋亡、活性氧(ROS)水平、Collagen Ⅱ、MMP-3、MMP-13及AMPK/SIRT1/NF-κB通路相关蛋白表达.结果:白细胞介素-1β组较对照组克隆形成数量、SOD水平、Collagen Ⅱ、SIRT1表达、p-AMPK/AMPK比值降低,细胞凋亡率、IFN-γ、IL-6、TNF-α、MDA水平、荧光强度、MMP-3、MMP-13表达、Ac-NF-κB p65/NF-κB p65比值升高,差异有统计学意义(P<0.05);低/中/高剂量橘红素组较白细胞介素-1β组克隆形成数量、SOD水平、Collagen Ⅱ、SIRT1表达、p-AMPK/AMPK比值升高,细胞凋亡率、IFN-γ、IL-6、TNF-α、MDA水平、荧光强度、MMP-3、MMP-13表达、Ac-NF-κB p65/NF-κB p65比值降低,差异有统计学意义(P<0.05);高剂量橘红素+Compound C组较高剂量橘红素组克隆形成数量、SOD水平、Collagen Ⅱ、SIRT1表达、p-AMPK/AMPK比值降低,细胞凋亡率、IFN-γ、IL-6、TNF-α、MDA水平、荧光强度、MMP-3、MMP-13表达、Ac-NF-κB p65/NF-κB p65比值升高,差异有统计学意义(P<0.05).结论:橘红素对IL-1β诱导的软骨细胞损伤的改善作用可能通过激活AMPK/SIRT1/NF-κB通路实现.

Objective:To investigate the effect of tangeretin on interleukin-1β(IL-1β)-induced chondrocyte injury and ana-lyze its mechanistic association with the AMP-activated protein kinase(AMPK)/silent information regulator 2 homolog 1(SIRT1)/nuclear factor κB(NF-κB)signaling pathway.Methods:Chondrocytes were cultured and divided into control group,interleukin-1β group,low-dose,medium-dose,and high-dose tangeretin groups,and high-dose tangeretin plus AMPK inhibitor Compound C group.Cloning formation assays,flow cytometry,DCFH-DA fluorescent probes,and West-ern Blot were used to detect cell proliferation,apoptosis,ROS levels,Collagen Ⅱ,MMP-3,MMP-13,and the expression of proteins related to the AMPK/SIRT1/NF-κB pathway.Results:The interleukin-1β group showed reduced clone formation number,SOD levels,Collagen Ⅱ,SIRT1 expression,p-AMPK/AMPK ratio,as well as increased apoptosis rate,IFN-γ,IL-6,TNF-α,MDA levels,fluorescence intensity,MMP-3,MMP-13 expression,and Ac-NF-κB p65/NF-κB p65 ratio compared to the control group(P<0.05).The low-dose,medium-dose,and high-dose tangeretin groups showed increased clone for-mation number,SOD levels,Collagen Ⅱ,SIRT1 expression,p-AMPK/AMPK ratio,as well as reduced apoptosis rate,IFN-γ,IL-6,TNF-α,MDA levels,fluorescence intensity,MMP-3,MMP-13 expression,and Ac-NF-κB p65/NF-κB p65 ratio compared with the interleukin-1β group(P<0.05).The high-dose tangeretin+Compound C group showed re-duced clone formation number,SOD level,Collagen Ⅱ,SIRT1 expression,p-AMPK/AMPK ratio,increased apoptosis rate,IFN-γ,IL-6,TNF-α,MDA levels,fluorescence intensity,MMP-3,MMP-13 expression,and Ac-NF-κB p65/NF-κB p65 ratio compared to the high dose tangeretin group(P<0.05).Conclusion:The improvement effect of tangeretin on IL-1β-induced chondrocyte damage may be achieved by activating the AMPK/SIRT1/NF-κB signaling pathway.
